人食管鳞癌p75NTR的核阳性表达细胞特性的研究
Study on the characteristics of the esophageal cancer cells expressing the p75NTR in the nucleus in human esophageal squamous cell carcinoma
【摘要】 目的研究人食管癌Eca109细胞中p75NTR的核阳性表达细胞的特性。方法运用免疫组织化学技术检测食管鳞癌标本中p75NTR的核阳性表达情况并分析与各种预后因子的相关性。运用无血清成球培养法培养Eca109细胞,富集到第1代肿瘤干细胞球样细胞,并进行传代培养,得到第1代、3代、5代、7代和9代细胞球样细胞。检测不同代次细胞球细胞的细胞周期及成瘤情况,并进行对比。结果 p75NTR的核阳性表达与年龄、肿瘤直径和病理分级有关,而与其他相关因素(比如:性别、淋巴结转移等)无显著相关性。第1代、3代、5代细胞球细胞处于G0/G1期细胞比例随细胞球细胞代数的增长依次递增,成瘤能力依次增强;第5代、第7代和第9代细胞球细胞处于G0/G1期细胞比例没有明显变化。结论食管癌p75NTR核阳性细胞可能是食管癌干细胞。
【Abstract】 Objective We have found that the cells expressing p75NTR in the nucleus in human esophageal cancer cell lines had the chacteristics of cancer stem cells. In this to further prove the cancer stem cell characteristics of the cells expressing p75NTR in the nucleus in human esophageal cancer cells. Methods The p75NTR expression in esophageal squamous cell carcinoma specimens was detected by immunohistochemistry and the correlation with various prognostic factors was analyzed. Eca109 cells were cultured in serum-free pellet culture medium to enriched the first generation of tumor stem cells,and then resuspended the cell pellets to get the third,fifth,seventh and ninth generation of cells with the same culture medium. The cell cycle and tumorigenesis of different generation pellets cells were measured. Results The number of cell expressingp75NTR in the nucleu was associated with the age of the patient,the tumor diameter and the pathological grade of esophageal cancer,but had no significant correlation with other related factors( such as gender,lymph node metastasis,etc.). The proportion of cells in G0/G1 phase and the tumorigenic ability graduatly increased with the growth of the pellets generation in the first,third and fifth generation cell pellets. There was no significant change in the proportion of cells in G0/G1 phase in the5 th,7 th and 9 th generations. Conclusion The cells expressing p75NTR in the nucleu are the stem cells of esophageal carcinoma.
