脂联素对人髓核细胞增殖凋亡的调控作用
The Regulation of Adiponectin on Human Nucleus Pulposus Cells Proliferation and Apoptosis
【摘要】 目的探讨脂联素在人髓核细胞增殖和凋亡中发挥的作用,并且研究其初步机制。方法提取并培养人髓核细胞、构建脂联素受体慢病毒(LV-sh Adipo R1和LV-sh Adipo R2)和对照空载体病毒,将病毒转染至人髓核细胞。肿瘤坏死因子α(tumor necrosis factor-alpha,TNF-α)诱导髓核细胞凋亡模型,采用流式细胞仪、TUNEL实验和CCK8试剂盒检测脂联素调控髓核细胞增殖凋亡的作用。结果 TNF-α能抑制髓核细胞增殖并诱导其凋亡,脂联素能逆转TNF-α对髓核细胞增殖和凋亡的调控作用(P<0.05)。Adipo R1/2沉默后,髓核细胞增殖受到抑制、凋亡升高,以Adipo R2为著(P<0.05)。结论脂联素能够通过两个受体(Adipo R1、Adipo R2)调控髓核细胞增殖和凋亡。
【Abstract】 Objective To investigate the role of adiponectin on human nucleus pulposus cells proliferation and apoptosis.Methods TNF-αwas used to induce apoptosis in human nucleus pulposus cells.CCK8 test,TUNEL and flow cytometry were used to detect the proliferation and apoptosis of human nucleus pulposus cells.Gain and loss of function was used to investigate the role and mechanism of adiponectin on the proliferation and apoptosis of human nucleus pulposus cells.Results TNF-αinhibited proliferation and induced apoptosis of human nucleus pulposus cells,adiponectin could antagonize the proliferation and apoptosis induction in human nucleus pulposus cells by TNF-α.Silence of Adipo R1/R2 can enhance human nucleus pulposus cells proliferation and apoptosis induced by TNF-α.Conclusion Adiponectin can regulate the proliferation and apoptosis of human nucleus pulposus cells through Adipo R1 and Adipo R2.
