年龄、BMI、吸烟和饮酒对男性精子印记基因CpG位点甲基化的影响
Effects of age, BMI, smoking and drinking on methylation status of CpG sites of imprinted genes in male sperm
【摘要】 目的观察不同人口学特征的男性精子印记基因甲基化水平的分布情况。方法收集440例进行孕前检查已婚男性的一般资料和精液样本。提取精子DNA并进行亚硫酸盐修饰,采用焦磷酸测序法对精子H19、Peg3、Meg3基因甲基化差异区(DMR)CpGs位点的甲基化水平进行检测。比较不同年龄、BMI、吸烟和饮酒情况者精子印记基因的甲基化水平。结果不同年龄、BMI、饮酒情况者H19、Peg3、Meg3基因DMR各CpG位点甲基化水平差异无统计学意义。吸烟者H19基因的CpG2、CpG7位点和Meg3基因的CpG2位点甲基化水平高于非吸烟者(P均<0.05)。结论不同年龄、BMI和饮酒情况的男性精子H19、Peg3、Meg3基因甲基化水平无统计学差异;有吸烟习惯的男性精子H19基因的CpG2、CpG7位点和Meg3基因的CpG2位点甲基化水平高于非吸烟者。
【Abstract】 Objective To observe the distribution of methylation levels of imprinted genes in male sperm with different demographic characteristics. Methods The general data and semen samples of 440 married men were collected. Sperm DNA was extracted, and modified with sulfites. The methylation levels of CpGs sites in differentially methylated region(DMR) of H19, Peg3 and Meg3 genes were detected by pyrosequencing. Then, we compared the methylation levels of sperm-imprinted genes in men of different age, BMI, smoking, and drinking status. Results There were no significant differences in the methylation levels of CpG sites in DMR of H19, Peg3, and Meg3 between men of different age, BMI, and drinking status. However, the methylation levels of CpG2 and CpG7 sites of H19, and CpG2 site of Meg3 in smokers were higher than those in non-smokers(all P<0.05). Conclusion There were no significant differences in methylation levels of H19, Peg3 and Meg3 of male sperm between men of different age, BMI and drinking status. The methylation levels of CpG2 and CpG7 sites of H19, and CpG2 site of Meg3 are higher in smokers than in non-smokers.
