499例手足口病患儿病原谱与分子流行病学特征分析
Pathogen spectrum and molecular characteristics of 499 children with hand-foot-and-mouth disease
【摘要】 目的对499例手足口病(HFMD)患儿的病原谱与分子流行病学特征进行分析。方法采集2014~2016年天津市儿童医院收治的HFMD患儿粪便标本499份,采用RT-PCR法检测肠道病毒(EV)阳性率,进一步采用RT-PCR法对EV阳性标本进行衣壳蛋白VP1(VP1)区全长基因扩增和序列检测,用生物软件Chromas 1. 62及MEGA6. 06分析部分标本测序结果并构建系统进化树。结果 499份标本中检出EV阳性429份,EV71、柯萨奇病毒A组16型(CVA16)及非EV71非CVA16其他EV分别占54. 1%、6. 3%、39. 6%; 170份其他EV阳性标本中检出CVA10阳性47份、CVA6阳性35份。VP1亲缘进化树分析结果显示,28份EV71序列均为C4基因亚型的C4a分支,22份CVA10序列均为G型,16份CVA6序列均为F型。结论 499例HFMD患儿中共检出21种EV,EV71的阳性率最高,非EV71非CVA16的其他EV类型感染比例也较高。
【Abstract】 Objective To analyze the pathogen spectrum and molecular characteristics of 499 children with hand-footand-mouth disease( HFMD). Methods All 499 samples of HFMD children collected from Tianjin Children’s Hospital from2014 to 2016 were subsequently tested for the virus nucleotide acid type by RT-PCR. We used RT-PCR to perform full-length gene amplification and sequence detection of the capsid protein VP1( VP1) region in enterovirus( EV)-positive samples,and used the bio-software Chromas 1. 62 and MEGA6. 06 to analyze the sequencing results and construct a phylogenetic tree. Results All the 499 HFMD,429 EV positive cases were found,the constituent ratio of EV71,CVA16 and other EV were54. 1%,6. 3% and 39. 6%,respectively. In 170 samples of other EV,there were 47 cases of CVA10 and 35 cases of CVA6.Phylogenetic analysis showed that EV71 belonged to genotype C4 a,CVA10 belonged to genotype G and CVA6 belonged to genotype F,respectively. Conclusion Twenty-one EV were detected in this study,with the highest positive rate of EV71,and the types and proportions of other non-EV71 and non-CVA16 enteroviruses show the trend of increasing.
