基质金属蛋白酶-9对奶牛乳腺上皮细胞泌乳的影响
Effect of matrix metalloproteinase 9 on lactation of dairy cow mammary gland epithelial cells
【摘要】 利用基质金属蛋白酶-9(matrix metalloprotein 9,MMP9)过表达载体或干扰片段转染奶牛乳腺上皮细胞后,分别用荧光定量PCR(Quantitative real-time PCR,q PCR)、甘油三酯试剂盒和MTT细胞增殖检测试剂盒检测奶牛乳腺上皮细胞的β-酪蛋白表达、乳脂含量和增殖情况。结果表明,与对照组相比,MMP9过表达能显著提高奶牛乳腺上皮细胞中的β-酪蛋白表达和甘油三酯含量,促进细胞增殖;MMP9抑制时则获得相反结果。MMP9能促进奶牛乳腺上皮细胞泌乳。
【Abstract】 The overexpression vector or interference fragment of Matrix Metalloprotein 9(MMP9) was transfected into dairy cow mammary gland epithelial cells. Quantitative real-time PCR was used to detect mRNA expression of β-casein. Triglycerides assay kit was used to determine the content of milk fat. MTT assay was used to measure the cell proliferation.The results showed that dairy cow mammary gland epithelial cells with overexpressed MMP9 increased β-casein mRNA level and triacylglycerol levels, and enhanced the cell proliferation, compared with the control group. Inhibition of MMP9 produced the opposite effects. Conclusion: MMP9 can promote lactation in dairy cow mammary gland epithelial cells.
