层粘连蛋白对奶牛乳腺上皮细胞乳蛋白表达的调控
Regulation of laminin on milk protein expression in cow mammary epithelial cells
【摘要】 激素或生长因子调控依赖于整联蛋白介导的细胞-细胞外基质相互作用,层粘连蛋白及其细胞表面特异受体整联蛋白β1亚基是小鼠泌乳分化的必要条件,但它们参与奶牛泌乳分化调控的作用机制尚待阐明。本研究以LN作为培养底物,体外培养泌乳期奶牛乳腺上皮细胞,添加催乳复合物诱导BMECs的乳蛋白表达,并对整联蛋白的β1亚基进行了组织和细胞定位,实验结果显示,添加LN能在蛋白水平增加整联蛋白β1亚基表达,同时对催乳复合物诱导乳铁蛋白和β-酪蛋白表达也有促进作用,因此,上调整联蛋白β1表达来增强BMECs对催乳素的应答可能是LN促进泌乳分化的一种作用方式。
【Abstract】 The regulation of hormones or growth factors dependent upon integrin-mediated cell-extracellular matrix interactions, required laminin(LN) and β1 integrin as its specific receptor on cell surface. However, the mechanism of lactation differentiation in bovine mammary epithelial cells(BMECs) is unclear. In this study, LN as a culture substrate, lactogenic complex were added to induce milk protein expression in BMECs, and β1 integrin were detected. The results showed that LN increased integrin β1 expression at the protein level, and also promoted the expression of lactoferrin and β-casein induced by lactogenic complex in BMECs. To induce a higher integrin β1 expression level to enhance the response of BMECs for prolactin may be a mode of action for LN to promote lactation differentiation.
