小鼠骨髓来源巨噬细胞内多肽的分离鉴定
Isolation and identification of polypeptides from murine bone marrow derived macrophages
【摘要】 以小鼠骨髓来源巨噬细胞(BMDM)作为研究对象,分离鉴定巨噬细胞内的多肽。将获得的小鼠骨髓来源巨噬细胞分两组,实验组细胞用LPS刺激,对照组用PBS处理,共培养4 h,超声破碎、过10 ku超滤管后,采用液-质联用仪鉴定分析细胞内的多肽,并将得到的多肽序列与报道过的多肽序列进行比对,确定新发现的多肽。结果表明:发现由8~29氨基酸组成的、来源于小鼠骨髓巨噬细胞内多肽340条,其中102条是从未公开报道过。本研究发现的多肽,旨为开展动物体内具有特定功能的生物活性多肽的研究提供借鉴。
【Abstract】 The bone marrow-derived macrophages(BMDM)of mouse were used as research objects to isolate and identify polypeptides in macrophages. Methods:BMDM were divided into two groups. The experimental group cells were stimulated with LPS, the control groupwas treated with PBS, cultured for 4 h, sonicated, and after passing 10 ku ultrafiltration tube, The polypeptide within the cell is identifiedandanalyzed by liquid-mass spectrometry, and the resulting polypeptide sequence is aligned with the reported polypeptide sequence to determinethe newly discovered polypeptide.Result:A total of 340 polypeptides consisting of 8~29 amino acids were derived from mouse bone marrowmacrophages. Of these, 102 were polypeptides that had never been reported before and were discovered in this study. They are organisms that have specific functions in animals. The foundation for the study of active peptides.
