基于便秘模型小鼠相关差异蛋白表达探讨中医“肺与大肠相表里”理论

【摘要】 【目的】从"肠病及肺"角度,研究便秘模型小鼠肺组织差异表达蛋白质,寻找肺与大肠的病理联系,为阐明中医"肺与大肠相表里"理论实质提供物质基础。【方法】运用蛋白质组学技术,对正常对照组和便秘模型组小鼠肺组织总蛋白质进行双向凝胶电泳(2-DE),选择差异蛋白质斑点,进行基质辅助激光解吸电离—串联飞行时间质谱(MALDI-TOF/TOF)分析和生物信息学分析。【结果】筛选出3个差异表达蛋白质点,其中细胞色素c氧化酶亚基5A(COX5A)、过氧化物氧化还原酶2(Prdx-2)蛋白在便秘组表达下调,肌球蛋白调节轻链2(MLC-2)蛋白表达上调。【结论】便秘小鼠"肠病及肺"的机制可能与肺细胞发生氧化应激损伤有关。

【Abstract】 Objective To research the differential proteins expression in lung tissues of constipation mouse model characterized as large intestine disease involving the lung,and to discuss the pathological correlation between the lung and large intestine,thus to further provide the evidence for the theory of the lung and large intestine being interiorly and exteriorly related.Methods Total proteins in lung tissues of normal control group and constipation model group were measured by proteomic method and two-dimensional gel electrophoresis(2-DE).And then the differential proteins were screened and identified by matrix-assisted laser desorption/ionization(MALDI)time-offlight/time-of-flight(TOF/TOF).Results Three differential proteins were found.Of the 3 proteins,cytochrome c oxidase subunit 5A(COX5A)and peroxiredoxin-2(Prdx-2)were down-regulated,and myosin regulatory light chain 2(MLC-2)were up-regulated.Conclusion The pathogenesis for large intestine disease involving the lung in constipation mice may be related with oxidative stress in lung tissues.
