不同提取方法对银杏叶多糖含量的影响
Effects of Extraction Methods on Content of Polysaccharide from Folium Ginkgo
【摘要】 【目的】分析不同提取方法对银杏叶中多糖含量的影响,为其进一步开发应用提供依据。【方法】采用水提醇沉法、酶解—水提醇沉法、超声波—水提醇沉法、微波—水提醇沉法制备银杏叶多糖,以称质量法测定提取物得率,同时以苯酚—硫酸法进行显色后通过紫外—可见分光光度计测定多糖含量。【结果】不同提取方法的银杏叶多糖提取物得率及含量分别为:水提醇沉法,12.58%、5.69%;酶法—水提醇沉法,14.05%、6.68%;超声波—水提醇沉法,14.12%、6.82%;微波—水提醇沉法,13.31%、6.15%。其中,含量比较具有显著性差异(P <0.05或P <0.01)。【结论】不同提取方法制备的银杏叶多糖提取物得率和含量不一,其中以超声波—水提醇沉法最高、水提醇沉法最低。
【Abstract】 Objective To analyze the influence of different extraction methods on the content of polysaccharide from Folium Ginkgo,thus to provide the reference for the further development and application of Folium Ginkgo.Methods The water-extraction alcohol-precipitation method, cellulose enzyme-assisted extraction method,ultrasound-assisted extraction method, and microwave-assisted extraction method were adopted to isolate polysaccharide from Folium Ginkgo. The yield of extracts was determined by weighing method,and the content of polysaccharide was measured by phenol-sulfuric acid method combined with ultraviolet spectrophotometry. Results The extraction yield and content of polysaccharide from Folium Ginkgo by different extraction methods were12.58% and 5.69% by the water-extraction alcohol-precipitation method,14.05% and 6.68% by the cellulose enzyme-assisted extraction method,14.12% and 6.82% by the ultrasound-assisted extraction method,13.31%and 6.15% by the microwave-assisted extraction method,respectively. There were significant differences in the contents(P < 0.05 or P < 0.01). Conclusion The yield of extracts and polysaccharide content in Folium Ginkgo by different extraction methods are different,by which the ultrasound-assisted extraction method is the highest and the water-extraction alcohol-precipitation method is the lowest.
