二氟甲基鸟氨酸对2型糖尿病大鼠心肌细胞凋亡的影响
Effects of difluorometylornithine on cardiomyocyte apoptosis in type 2 diabetic rats
【摘要】 目的研究二氟甲基鸟氨酸(difluoromethylornithine,DFMO)对2型糖尿病(type 2 diabetic melitus,T2DM)大鼠心肌细胞凋亡及对凋亡相关蛋白Caspase-3的影响。方法采用高糖高脂膳食负荷链脲佐菌素(streptozotocin,STZ)单次腹腔注射建立T2DM大鼠模型。将50只实验大鼠分为正常对照组,糖尿病组和DFMO干预组三组。12周后检测各组大鼠空腹血糖、心脏参数、心肌细胞凋亡和Caspase-3蛋白表达。结果 DFMO治疗后降低糖尿病大鼠空腹血糖和心脏参数(P<0.05),心肌细胞凋亡程度下降(P<0.05)。同时,DFMO干预下调Caspase-3蛋白表达(P<0.05)。结论 DFMO抑制T2DM大鼠心肌细胞凋亡,机制与下调Caspase-3蛋白表达有关。
【Abstract】 Objective To investigate the influence of difluorometylornithine( DFMO) on cardiomyocyte apoptosis and the protein expression of Caspase-3 in type 2 diabetic mellitus( T2 DM) rats.Methods The type 2 diabetic mellitus rat model was induced by 4 weeks ’ high sucrose-high fat diet feeding and one-time intraperitoneal injection of streptozotocin.Rats were randomly divided into 3 groups: control group,diabetes model group and DFMO treatment group.Rats of treatment group were treated with 2% DFMO drinking for 12 weeks.The changes in blood glucose,cardiac index,cardiomyocyte apoptosis and Caspase-3 protein expression were calculated.Results Compared with diabetes model group,the blood glucose,the index of cardiomyocyte apoptosis was decreased,meanwhile,the protein expression of Caspase-3 was decreased in treatment group( P < 0. 05).Conclusions The results indicate that DFMO exerts protective effects on cardiomyocyte apoptosis in T2 DM rats which may be related to its function on the down regulate protein expression of Caspase-3.
