基于金属荧光增强效应检测ATP的研究

【摘要】 利用银纳米粒子的金属荧光增强效应,构建荧光传感器实现对小分子ATP的检测。首先将修饰荧光基团的ATP适体固载在银纳米粒子表面,通过杂交互补将标记猝灭基团的DNA与之结合,实现荧光共振能量转移降低背景信号。ATP与适体链的特异性结合破坏荧光共振能量转移,荧光基团靠近银纳米粒子表面,由于银纳米粒子的金属荧光增强效应,使荧光信号增强,实现对ATP的检测,此种方法也可以用于其他生物小分子、蛋白质等的检测。

【Abstract】 The silver nanoparticles(AgNPs)-based fluorescence resonance energy transfer(FRET) was reported for ATP detection. The fluorophore-functionalized ATP aptamer was first immobilized on the surface of the silver nanoparticles. Then, the quencher-carrying strands were hybridized with the aptamer to form FRET senor. When the senor interacted with ATP, the FRET effect was disrupted and the fluorescence intensity was enhanced by AgNPs. This method had good selectivity and had been successfully applied to the analysis of ATP in human serum samples. The method has potential application value in biological detection with wide application range, strong selectivity and convenient operation.
