高碘对大鼠甲状腺血清激素水平及NIS基因表达的影响

【摘要】 目的观察高碘对大鼠甲状腺血清激素水平及NIS基因表达的影响。方法选取健康Wistar大鼠40只,随即分成4组,分别为正常组(NI)、10倍高碘组(10HI)、50倍高碘组(50HI)、100倍高碘组(100HI)各10只,饲养3个月后称取甲状腺质量,检测血清甲状腺激素水平和甲状腺组织中NIS mRNA表达水平。结果各HI组大鼠甲状腺相对质量与绝对质量与NI组比较差异无统计学意义(P>0.05);10HI组、50HI组血清激素TT4、FT4、TT3水平与NI组比较差异无统计学意义(P>0.05);100HI组血清激素TT4、FT4、TT3水平均低于NI组,50HI组和100HI组FT3水平均低于NI组,差异均有统计学意义(P<0.05);各HI组与NI组NIS mRNA表达水平比较有所下降,但差异无统计学意义(P>0.05)。结论过量的碘摄入可使大鼠血清甲状腺激素水平和NIS mRNA的表达水平发生变化,从而可能引发某些甲状腺疾病。

【Abstract】 Objective To investigate the effects of high iodine on serum hormone level and NIS gene expression in rats thyroid.Methods Forty healthy Wistar rats were randomly divided into 4 groups:normal group(NI),10 times high iodine group(10 HI),50 times high iodine group(50 HI),100 times high iodine group(100 HI),each of 10 rats.The thyroid weight was measured after feeding for 3 months,the levels of serum thyroid hormone and the expression of NIS mRNA in thyroid tissue were detected.Results There was no significant difference in the relative mass and absolute mass of thyroid between each HI group and NI group(P>0.05).There were no significant differences in serum hormones TT4,FT4,TT3 between 10 HI group and 50 HI group compared with NI group(P>0.05).The serum levels of TT4,FT4 and TT3 in 100 HI group were lower than those in NI group;The FT3 levels in 50 HI group and 100 HI group were lower than those in NI group,the difference was statistically significant(P<0.05).The expression levels of NIS mRNA in each HI group and NI group were decreased,but the difference was not statistically significant(P>0.05).Conclusion Excessive iodine intake can change the level of serum thyroid hormone and the expression of NIS mRNA in rats,which may lead to the occurrence of certain thyroid diseases.
