HPLC-DAD-ELSD测定不同产地黄芪中2种成分的含量
Determination of two constituents in Radix Astragali from different places by HPLC-DAD-ELSD
【摘要】 目的:采用HPLC-DAD-ELSD同时测定不同产地黄芪中黄芪甲苷、芒柄花素含量。方法:采用HPLC-DAD-ELSD串联技术,检测波长:265nm;流动相:A水,B乙腈;流速:1mL·min-1;柱温:35℃;进样量:20μL;增益值:20;漂移管温度:60℃;加热器级别:60%,载气压力:35PSI。结果:黄芪甲苷在0.1~1.6mg·mL-1,峰面积对数和浓度对数呈良好的线性关系;芒柄花素在0.02~0.32mg·mL-1,峰面积和浓度呈良好的线性关系,相关系数分别为0.9997、0.9999,各方法学指标和加样回收率均符合要求。结论:本方法重复性良好,结果准确可靠,可用于黄芪中两种不同类型成分的含量测定。
【Abstract】 Objective: To determine the contents of astragaloside IV and formononetin in Radix Astragali from different places by HPLC-DAD-ELSD. Method: HPLC-DAD-ELSD were applied in series and detected at 265 nm. The mobile phase was consisted of A(water)and B(acetonitrile),flow rate was 1 mL·min-1. Column temperature: 35℃,sample size: 20μL,gain: 20,tube: 60℃,neb: 60%,air pressure:35 PSI. Results: The calibration curves of astragaloside IV and formononetin were linear in the range of 0.1 ~1.6 mg·mL-1 and 0.02 ~0.32 mg·mL-1,the correlation coefficient were 0.9997 and 0.9999,the technology indexes and recoveries were both qualified. Conclusion:It is concluded that the method is accurate and reliable to evaluate the contents of two different constituents in Radix Astragali. The re-producibility is well.
