棉铃虫幼虫血淋巴免疫因子溶血素的理化特性研究
Study on physicochemical properties of hemolysin(s) in Helicoverpa armigera larval hemolymph
【摘要】 为深入研究棉铃虫幼虫血淋巴免疫因子溶血素的分子结构和生理功能,明确其分离、纯化的理化条件,采用分光光度法分别研究了温度、浓度、作用时间、pH、EDTA和金属离子对该溶血素的溶血活性影响及其糖结合特性。结果表明,棉铃虫幼虫血淋巴溶血素属热不稳定型。溶血素反应浓度与溶血活性并不成正比,8倍稀释液的溶血活性反高于4倍稀释液。反应2 h后,血淋巴溶血素对鸡血红细胞溶解速度急剧加快,6 h溶血效能达最大。血淋巴溶血活性的最适pH 7.0~8.0。62.5~500 mmol·L-1EDTA对血淋巴溶血活性具有促进作用,而Ca2+却具有极显著的抑制作用。糖抑制试验结果表明,供试的8种糖(糖苷)和唾液酸均未对血淋巴溶血活性产生抑制作用。
【Abstract】 In order to explore deeply the molecular structure and physiological function of the immune-related factor hemolysin(s) found in Helicoverpa armigera larval hemolymph,and to make sure its physicochemical conditions of separation and purification,the effects of temperature,concentration,action time,pH,EDTA and metal ions on the hemolytic activity of hemolysin(s) and carbohydrate-binding properties were respectively researched by using the spectrophotometric method.The results showed that the hemolysin(s) displayed thermolability.The concentrations of hemolymph hemolysin(s) were not in direct proportion to their hemolytic activity.The hemolytic activity of 8-fold diluent was higher than that of 4-fold diluent.At 2 h after hemolymph and erythrocytes were in the action,the rate of erythrocytes lysis increased dramatically,and complete hemolysis reached at 6 h after the action.The optimum pH for hemolymph hemolytic activity was at 7.0~8.0.The concentration of EDTA from 62.5 to 500 mmol·L-1 could promote hemolytic activity of hemolymph,while the Ca2+ possessed an extremely significant inhibition effect.The result of carbohydrate-inhibition assay indicated that hemolytic activity of hemolymph could not be inhibited by the 8 carbohydrates or glycoside and sialic acid for test.
