MiR-21和MiR-137的表达对膀胱癌细胞增殖和侵袭的影响
Effects of MiR-21 and MiR-137 expression on proliferation and invasion of bladder cancer cells
【摘要】 目的:研究microRNA-21(MiR-21)和microRNA-137(MiR-137)在膀胱癌细胞的表达,分析其对细胞增殖和侵袭的影响。方法:选择2014年1月至2017年10月期间,在我院经病理检查确诊,并行手术切除治疗的96例膀胱癌患者为研究对象,通过PCR等检测技术,检测膀胱癌肿组织及癌肿周围组织(癌旁)内MiR-21和MiR-137的表达,研究MiR-21和MiR-137对膀胱癌细胞增殖和侵袭的影响。结果:膀胱癌肿组织MiR-21、MiR-137表达量显著高于癌肿周围组织,随着膀胱癌恶性度的增加而增加;癌肿细胞内TRAP-1、KPNA 2、TRPM 8的mRNA表达量高于癌旁组织,CBX 7、CDH 13低于癌旁组织;MiR-137和MiR-21高表达的膀胱癌细胞的24 h、48 h和72 h后增殖速率均明显增快;Transwell实验显示过表达组细胞过膜数量为(402±25/视野),高于空白对照组和抑制组(P<0.05)。结论:MiR-21和MiR-137的表达量与侵袭和增值能力相关,表达增多可显著提高膀胱癌细增殖和侵袭,可为临床上检查、诊断、治疗膀胱癌提供科学依据。
【Abstract】 Objective: The expression of microRNA-21(MiR-21) and microRNA-137(MiR-137) in bladder cancer cells was studied, and its effects on cell proliferation and invasion were analyzed. Methods: From January 2014 to October 2017, 96 patients with bladder cancer who were diagnosed with concurrent surgical resection by pathological examination in our hospital were selected as the subjects.Using PCR and other detection techniques, the expression of MiR-21 and MiR-137 in bladder cancer tissues and surrounding tissues(adjacent to cancer) were detected, and the effects of MiR-21 and MiR-137 on the proliferation and invasion of bladder cancer cells were investigated. Results: The expression levels of MiR-21 and MiR-137 in bladder cancer tissues were significantly higher than those in the surrounding tissues, and increased with the increase of malignancy of bladder cancer. The mRNA expression levels of trap-1, KPNA 2 and TRPM 8 in cancer cells were higher than that of adjacent tissues, and CBX 7 and CDH 13 were lower than those of the adjacent tissues. Both MiR-137 and MiR-21 high expression of bladder cancer cell proliferation rate were significantly increased after 24 h, 48 h and 72 h. Transwell experiment showed that the number of cells in the expression group was(402±25/field), higher than that in the blank control group and the suppression group, and the difference was statistically significant(P<0.05).Conclusion: The expression quantity of MiR-21 and MiR-137 is related to invasion and appreciation ability, and the increase of expression can significantly improve the proliferation and invasion of bladder cancer, which can provide scientific basis for clinical examination, diagnosis and treatment of bladder cancer.
