神州草SOD提取工艺研究

【摘要】 采用膜处理、脱色、混合有机溶剂除杂、丙酮沉淀等工艺提取纯化神州草中的SOD,通过考查浸提缓冲液、活性炭脱色时间、激活剂加量、混合溶剂及丙酮加量等因素对SOD酶活性的影响,确定神州草SOD的最佳提取纯化工艺为:在神州草叶中加入2.5倍pH值7.8的0.9%氯化钠溶液打浆、离心,上清液经截留分子量为100 000的中空纤维膜,透过液再经截留分子量为10 000的中空纤维膜浓缩后得到SOD浓缩。40 m L浓缩液中加入0.5 g活性炭和0.5 g活性白土及150μL氯化铜激活剂后脱色20 min,离心得上清液,加入总体积50%的氯仿-乙醇混合体系(3∶5),离心后,上清液中加入1.0~1.2倍体积的丙酮沉淀,得到SOD粗品。该粗品再经混合有机溶剂除杂、丙酮沉淀处理后,获得活性在2 000 U/mg·pro以上的SOD产品。为神州草的资源化综合利用提供参考。

【Abstract】 Membrane treatment, decolorization, removal foreign material of mixed organic solvent and acetone precipitation were applied to extract and purify SOD from Shenzhou grass in this study. The influences of leaching solution type,decolorization time of activated carbon, activated agent dosage, chloroform ∶ethanol mixture(3 ∶5) mixing system and acetone amount on SOD enzyme activity were investigated,and the results showed the optimum extraction and purification techniques of SOD from Shenzhou grass were that 1 kg Shenzhou grass with 2.5 times pH 7.8, 0.9% sodium chloride solution mixed,beating,after centrifugation to obtain a supernatant. The supernatant fluid was concentrated by a hollow fiber membrane with a molecular weight of 100 000, the permeated liquid was concentrated by hollow fiber membrane with a molecular weight of10 000 to obtain SOD concentrate. Putting 0.5~2.0 g of activated carbon and activated clay and 150 μL of copper chloride activator to 40 m L concentrated solution. Then the supernatant fluid was obtained by decolorization for 20 min and centrifugation.After centrifugation,putting the total volume of 50% chloroform∶ethanol mixture(3∶5) and 1.0~1.2 times the volume of acetone precipitation in the supernatant fluid to obtain SOD crude. The crude product was treated with organic solvent and acetone precipitation to obtain SOD products whose activity could reach to 2 000 U/mg·pro. This study provided a reference for the comprehensive utilization of Shenzhou grass.
