蛴螬多糖的一般药理学及亚急性毒性研究

【摘要】 目的:观察与评价蛴螬多糖对动物神经系统、呼吸系统、心血管系统的影响以及其亚急性毒性的作用特点,探讨其一般药理学作用和可能的副作用,为进一步开发与应用蛴螬多糖提供实验依据。方法:旷野法观察小鼠自发活动;睡眠时间法观察蛴螬多糖与戊巴比妥钠合用对小鼠的催眠协同作用;疲劳转棒实验观察机能协调活动;使用无创血压分析仪测定血压、心电图;对大鼠腹腔注射蛴螬多糖,14 d后进行脏器系数测定、血常规检查、血液生化指标检查以及组织病理学检查。结果:蛴螬多糖50、225、400 mg/kg腹腔注射,给药1次对小鼠自发活动无影响,给药1周对小鼠的机能协调活动无影响,与阈下剂量戊巴比妥钠无协同催眠作用;33、150、267 mg/kg腹腔注射1次,对大鼠呼吸频率无影响,但可略升高血压;亚急性毒性试验中,高剂量2周重复给药可明显增加血小板数量,降低清蛋白、高密度脂蛋白胆固醇的量,但以上指标均在正常范围内;对其它各项指标和组织结构等没有影响。结论:蛴螬多糖对呼吸系统、神经系统均无明显影响,对大鼠无明显毒性,是比较安全的蛴螬活性成分。

【Abstract】 Objective To observe and evaluate the effects of polysaccharide from Holotrichia diomphalia(PHD) on nervous system, respiratory system, cardiovascular system and its subacute toxicity in animals, explore its general pharmacological effects and possible side effects, and provide experimental evidence for further development and application of PHD. Methods The spontaneous activity of mice was observed in open filed. Hypnotic synergistic effect of PHD and pentobarbital sodium was observed on sleep time in mice. Coordination of function was observed by fatigue rotating rod test. Blood pressure and electrocardiogram were measured by noninvasive blood pressure analyzer. Organ coefficient measurement, blood biochemical index and histopathological examination were performed 14 days after intraperitoneally injection of PHD in rats. Results PHD(50, 225 and 400 mg/kg) was injected intraperitoneally once, and had no effect on the spontaneous activity of mice. One week after injection, PHD had no effect on the coordinated activity of mice, and had no synergistic hypnotic effect with the subthreshold dose of sodium pentobarbital. Intraperitoneal injection once at 33, 150 and 267 mg/kg had no effect on respiratory rate, but slightly increased blood pressure. In subacute toxicity test, repeated injection of high dose for 2 weeks could significantly increase the number of platelets and decrease the levels of albumin and high density lipoprotein cholesterol, but the above indexes were within the normal range. It had no effect on other indicators and organizational structure. Conclusion PHD has no obvious effect on respiratory system and nervous system, and has no obvious toxicity to rats. It is a safe active ingredient of Holotrichia diomphalia Bates.
