链脲佐菌素诱导1型糖尿病大鼠模型的最佳禁食时间与最优剂量
Optimal fasting time and dose of streptozotocin-induced diabetic rat model
【摘要】 目的通过观察不同禁食时间及不同剂量链脲佐菌素(STZ)对建立1型糖尿病(T1DM)大鼠模型的影响,探讨STZ建立T1DM动物模型的最佳禁食时间及最优剂量。方法选取清洁级雌性SD大鼠105只,随机分为7组,正常对照组(N组)及实验组(A、B、C、D、E、F组),每组15只。A组禁食12 h,STZ 50 mg/kg;B组禁食16 h,STZ 50 mg/kg;C组禁食20 h,STZ 50 mg/kg;D组禁食24 h,STZ 50 mg/kg;E组禁食20 h,STZ 60 mg/kg;F组禁食20 h,STZ 70 mg/kg;N组禁食20 h,等体积的柠檬酸盐缓冲液。注射STZ后第3天的随机血糖值>16.7 mmo1/L,并表现出明显的多饮、多食及多尿者判定为T1DM大鼠模型造模成功。结果 A组成模率60.00%,死亡率0;B组成模率66.70%,死亡率6.67%;C组成模率93.33%,死亡率6.67%;D组成模率93.33%,死亡率26.67%;E组成模率100.00%,死亡率26.67%;F组成模率93.33%,死亡率40.00%。结论禁食时间20 h,腹腔一次性注射STZ 50 mg/kg为建立T1DM大鼠模型的最佳禁食时间和最优剂量。
【Abstract】 Objective To investigate into the effects of different fasting times and different doses of streptozotocin(STZ) on the establishment of type 1 diabetes mellitus(T1DM) rat model, and to explore the best T1DM animal model established by STZ. Methods A total of 105 clean female SD rats were randomly selected and randomly divided into seven groups: the normal control group(N group) and experimental group(A,B, C, D, E, F groups), with 15 rats in each group. Fasting 12 h, STZ 50 mg/kg as group A; fasting 16 h, STZ50 mg/kg as group B; fasting 20 h, STZ 50 mg/kg as group C; fasting 24 h, STZ 50 mg/kg as group D; fasting20 h, STZ 60 mg/kg as group E; fasting 20 h, STZ 70 mg/kg as group F; fasting 20 h, equal volume citrate buffer solution as group N. Random blood glucose > 16.7 mmo1/L on the third day after STZ injection, showing obvious polydipsia; polyphagia and polyuria were judged as successful in the establishment of a rat model of type 1 diabetes. Results The compositional modulus rate was 60.00%, the mortality rate was 0 in group A; the compositional modulus rate was 66.70%, the mortality rate was 6.67% in group B; the compositional modulus rate was 93.33%, the mortality rate was 6.67% in group C; the compositional modulus rate was 93.33%,the mortality rate was 26.67% in group D; the compositional modulus rate was 100.00%, the mortality rate was 26.67% in group E; the compositional modulus rate was 93.33%, the mortality rate was 40.00% in group F. Conclusion Fasting time of 20 h and intraperitoneal injection of a STZ 50 mg/kg dose are good conditions for establishing the optimal fasting time and optimal dose for the T1DM rat model.
