大益菌方茶浓缩物的降血糖和降血脂作用研究
Study on Hypoglycemic and Hypolipidemic Effects of Dayi Bacterial Prescription Tea
【摘要】 目的通过建立小鼠高糖高脂动物模型,研究大益菌方茶浓缩提取物的降糖降脂作用。方法将220只健康雄性小鼠随机分成7组,分别为正常对照组(NC)、模型对照组(MC)、阳性对照组(PC)和观察组(大益菌方茶浓缩提取物DYA、DYB、DYC、DYD组);其中NC组和PC组为35只,其余组为30只。NC组给予正常基础饲料,PC组、MC组和观察组给予高糖高脂饲料。建模8周,从第9周开始,观察组灌胃给予大益菌方茶浓缩提取物,DYA组0.318 g·kg-1、DYB组0.300 g·kg-1、DYC组0.222 g·kg-1、DYD组0.298 g·kg-1,PC组灌胃给予83.33mg·kg-1二甲双胍和3.33 mg·kg-1洛伐他汀。分别于第15周处死动物,取血,分离血清,检测葡萄糖(Glu)、总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、总蛋白(TP)、白蛋白(ALB)、尿素和肌酐水平。结果与NC组相比,MC组血清Glu、TC和TG水平明显增高(P<0.01);与MC组相比,4个观察组血清Glu、TC和TG水平明显降低(P<0.01),HDL-C水平升高,血清胰岛素水平升高;与NC组和PC组相比,4个观察组血清Glu、TC和TG水平无明显差异。结论 4种大益菌方茶浓缩提取物能降低高糖高脂模型小鼠的血糖和血脂水平,实验效果与PC组作用相当。
【Abstract】 Objective To study the effects of concentrated extracts of 4 Dayi bacterial prescription tea for Hypoglycemic and Hypolipidemic by establishing a high-sugar and high-fat animal model in mice. Methods220 healthy male mice were randomly divided into 7 groups: normal group,positive control group,model group and experimental group(group A,B,C and D of concentrated extracts of Dayi bacterial prescription tea); 35 mice in normal group and positive control group,and 30 mice in other groups. The normal control group was fed with normal basic diet,while the other groups were fed with high-sugar and high-fat diet.Modeling for 8 weeks. Starting from the 9 th week,the experimental group was given concentrated extract of Dayi bacterial prescription tea by gavage administration. Group A(0.318 g·kg-1 DYA),Group B(0.3 g·kg-1 DYB),Group C(0. 222 g ·kg-1 DYC),Group D(0. 298 g ·kg-1 DYD),and the positive control group was given 83.33 mg·kg-1 metformin and 3.33 mg·kg-1 lovastatin by gavage administration. At the fifteenth week,the animals were killed,blood was taken,serum was separated,glucose(Glu),total cholesterol(TC),triglyceride(TG),high density lipoprotein cholesterol(HDL-C),alanine aminotransferase(ALT),aspartate aminotransferase(AST),total protein(TP), albumin(ALB), urea and Creatinine were detected. Results Compared with the normal control group,the levels of serum Glu,TC and TG in the model group increased significantly(P < 0. 05). Compared with the model group,the levels of serum Glu,TC and TG in the four experimental groups decreased significantly(P <0.05),while the levels of HDL-Cand insulin increased. Compared with the normal group and the positive drug group,the levels of serum Glu,TC and TG in the four experimental groups were increased significantly(P< 0. 05). There was no significant difference of the levels of serum Glu,TC and TG in the four experimental groups. Conclusion Concentrated extracts of 4 Dayi bacterial prescription tea can reduce blood sugar and lipid levels in high-sugar and high-fat model mice,and the experimental effect is similar to that of positive drugs.
