非小细胞肺癌组织中MNK1水平及临床意义

【摘要】 目的探讨非小细胞肺癌(NSCLC)组织中MAPK相互作用激酶1(MNK1)的表达及临床意义。方法收集本院2014年3月至2018年6月手术切除的NSCLC患者癌组织及配对癌旁组织79对,采用实时定量PCR检测上述组织的MNK1水平,分析MNK1水平与NSCLC临床病理特征的关系,采用Kaplan-Meier法分析MNK1水平与总生存期(OS)的关系,多因素分析采用Cox风险比例模型。结果 NSCLC组织的MNK1水平为4.668±1.264,高于癌旁组织的1.438±0.577(P<0.05)。MNK1表达与NSCLC的性别、年龄、吸烟史和病理类型均无关(P>0.05),而与肿瘤大小、淋巴结转移和TNM分期有关(P<0.05)。MNK1高表达者的中位OS为37.0个月,低表达者的中位OS未达,低表达者的中位OS长于高表达者(P<0.05);多因素分析显示MNK1表达是影响NSCLC预后的独立因素(OR=2.785,95%CI:1.506~5.941,P<0.05)。结论 MNK1在NSCLC组织中高表达,是NSCLC恶性程度高、预后差的指标,为NSCLC患者的早期诊断及预后评估提供指导。

【Abstract】 Objective To investigate the expression and clinical significance of MAPK interacting serine/threonine kinase 1(MNK1) in non-small cell lung cancer(NSCLC) tissues. Methods From March 2014 to June 2018, 79 pairs of cancer tissues and matched adjacent tissues of NSCLC patients were collected. The MNK1 level in the above tissues was detected by real-time quantitative PCR, and the relationship between MNK1 level and clinicopathological characteristics of NSCLC was analyzed. The relationship between MNK1 level and overall survival(OS) was analyzed by Kaplan-Meier method. Cox risk ratio model was used for multivariate analysis. Results The MNK1 level in NSCLC tissues was 4.668±1.264, higher than 1.438±0.577 in paracancer tissues(P<0.05). The expression of MNK1 was not related to sex, age, smoking history and pathological type of NSCLC(P>0.05), but related to tumor size, lymph node metastasis and TNM stage(P<0.05). The median OS of the patients with high expression of MNK1 was 37.0 months, and the median OS of the patients with low expression was not reached. The median OS of patients with low expression was longer than that of patients with high expression(P<0.05). Multivariate analysis showed that MNK1 expression was an independent factor affecting the prognosis of NSCLC(OR=2.785, 95%CI: 1.506-5.941, P<0.05). Conclusion The high expression of MNK1 in NSCLC is an indicator of high malignancy and poor prognosis of NSCLC, which provides guidance for early diagnosis and prognosis evaluation of NSCLC patients.
