Th17细胞对调节性B细胞的分化和IL-10分泌的影响
Effects of Th17 cells on regulatory B cell differentiation and IL-10 secretion
【摘要】 目的:探讨Th17细胞对体外调节性B(Breg)细胞分化的调控作用。方法:分离小鼠脾脏单个核细胞,进一步分选初始T细胞,利用转化生长因子(TGF)-β和白细胞介素(IL)-6诱导其向Th17细胞分化后,与初始B细胞共培养,体外利用脂多糖(LPS)或抗-IgM抗体诱导初始B细胞向Breg分化。检测Th17细胞对CD19~+CD5~+CD1d~+和CD19~+CD11b~+Breg细胞分化的影响,以及对其分泌IL-10等细胞因子和免疫球蛋白的影响。结果:Th17细胞可抑制CD19~+CD5~+CD1d~+Breg细胞分化,并抑制IL-10的分泌,对TGF-β、免疫球蛋白IgG、IgM和IgA分泌并无明显影响。结论:Th17细胞体外可抑制Breg细胞分化和IL-10的分泌。
【Abstract】 Objective: To explore the role of Th17 cells on the differentiation of regulatory B(Breg) cells in vitro. Methods:Mononuclear cells of spleens were isolated from mice. Following sorting, naive T cells were cultured with transforming growth factor(TGF)-β and interleukin(IL)-6 to induce their differentiation into Th17 cells. Afterward, Th17 cells were co-cultured with naive B cells. Naive B cells were differentiated into Breg cells in vitro by stimulation with lipopolysaccharide(LPS) or anti-IgM antibody. The effects of Th17 cells on the differentiation of CD19~+CD5~+CD1 d~+and CD19~+CD11 b~+Breg cells, and their secretion of IL-10, TGF-β, and immunoglobulins were analyzed. Results: Th17 cells inhibited the differentiation of CD19~+CD5~+CD1 d~+Breg cells and the secretion of IL-10, with no significant effects on the secretion of TGF-β, immunoglobulin IgG, IgM and IgA. Conclusion: Th17 cells can inhibit the differentiation of Breg cells and their secretion of IL-10 in vitro.
