骨桥蛋白对巨噬细胞破骨分化相关因子表达的影响

【摘要】 目的:探讨外源性骨桥蛋白(osteopontin,OPN)在体外对巨噬细胞破骨分化相关因子表达的影响。方法:体外培养小鼠RAW264. 7细胞,分为对照组(不加任何细胞因子),实验各组分别加入不同浓度的OPN(0. 1,0. 5,1. 0μg/m L)。培养48 h后qRT-PCR检测融合因子ATP6V0D2及破骨分化相关因子TRAP、MMP-9、CTSK的mRNA表达情况。结果:qRT-PCR显示,与对照组相比,OPN各组(0. 1,0. 5,1. 0μg/m L)的ATP6V0D2、TRAP、MMP-9、CTSK的mRNA表达均上调(P<0. 01)。结论:OPN能够促进巨噬细胞的破骨分化相关因子表达。

【Abstract】 Objective: To investigate the effect of osteopontin on the expression of osteoclast differentiation related factors in macrophages in vitro.Methods: Mouse RAW264. 7 macrophages were cultured in vitro,and then were induced as follows: set control group( without any cytokines),the experimental groups were treated with different concentrations of OPN( 0. 1,0. 5,1. 0 μg/m L). The macrophages were stimulated for 48 hours,and then the mRNA expression of fusion factor ATP6 V0 D2 and osteoclast related factors TRAP,MMP-9,CTSK were analyzed by qRT-PCR. Results: The qRT-PCR results showed that compared with the control group,ATP6 V0 D2,TRAP,MMP-9,CTSK expression increased significantly in all OPN groups( P<0. 01).Conclusion: OPN could promote the expression of osteoclast differentiation related factors in macrophages.
