前列腺癌中miR-612调节能量代谢、细胞增殖和凋亡的研究

【摘要】 探讨miR-612在前列腺癌中调节能量代谢、细胞增殖和凋亡中的作用.通过用miR-612转染LNCaP细胞,测量葡萄糖消耗、乳酸产生、细胞增殖、细胞凋亡和糖酵解相关蛋白.采用定量PCR方法,通过测定葡萄糖摄取率,乳酸产生水平;通过MTT、Western blot和FACS方法,验证miR-612对前列腺癌细胞能量代谢、增殖和凋亡的影响.转染miR-612后,LNCaP细胞的葡萄糖消耗、乳酸产生和细胞增殖减少,LNCaP细胞可观察到更显著的早期凋亡.用miR-612转染后,糖酵解相关蛋白HK2和PKM2的表达降低.研究结果显示,miR-612在前列腺癌细胞系中的过表达抑制了糖酵解和增殖,并增加了细胞凋亡.miR-612参与前列腺癌的糖酵解并发挥抑癌基因作用,并与前列腺癌的增殖和凋亡有关.

【Abstract】 The objective of our study is to investigate the role of miR-612 in regulating energy metabolism, proliferation and apoptosis in prostate cancer. By transfecting LNCaP cells with miR-612, glucose consumption, lactate production, cell proliferation, cell apoptosis and glycolysis-related proteins were measured. Quantitative PCR was used to verify the effects of miR-612 on energy metabolism, proliferation and apoptosis of prostate cancer cells by measuring glucose uptake rate, lactate production level, MTT, Western blotting and FACS. Glucose consumption, lactate production and cell proliferation of LNCaP cells decreased after transfection with miR-612. More early apoptosis of LNCaP cells was observed after transfection with miR-612. After transfection with miR-612, expression of glycolysis-related proteins HK2 and PKM2 decreased. Functional analysis showed that overexpression of miR-612 in prostate cancer cell lines suppressed their glycolysis and growth and increased apoptosis. All the results indicate that miR-612 is involved in the regulation of prostate cancer energy metabolism and the role of tumor suppressor genes.
