SIRT1在被动吸烟SD大鼠部分器官中的表达
Expression of SIRT1 in some Organs of passively smoking SD rats
【摘要】 目的通过被动吸烟诱导并建造被动吸烟大鼠模型,研究该模型氧化应激时SIRT1蛋白在肝、肺、肾组织中的表达情况,为研究被动吸烟对人体的危害提供参考。方法将大鼠用随机数法分为实验组20只,对照组20只,采用方苏榕等人的研究方法并加以改进建造被动吸烟大鼠模型,使用免疫荧光检测大鼠的肺、肝、肾等器官中SIRT1蛋白是否表达,Western-Blot半定量分析该器官中SIRT1蛋白的表达含量。结果实验组大鼠肺中SIRT1蛋白的表达量比对照组大鼠肺(P<0.05)中SIRT1蛋白的表达低;实验组大鼠肝(P<0.01)、肾(P<0.05)中SIRT1蛋白的表达量比对照组表达高。结论 SIRT1蛋白在被动吸烟大鼠的肺组织中表达含量低,在肝、肾组织中表达高。
【Abstract】 Objective To induce and construct a passive smoking rat model by passive smoking, and to study the expression of SIRT1 protein in liver, lung and kidney tissues during oxidative stress, and to provide a reference for studying the harm of passive smoking to human body. Methods Rats were randomly divided into the experimental group(n=20) and the control group(n=20). The passive smoking rat model was established by using the research method of Fang Surong and others. Immunofluoresence assay and western-blot was used to quantify were employed to qualitatively and quantitatively study the expression of SIRT1 protein in liver, lung and renal in smoked rats. Results The expression of SIRT1 protein in the lungs of the experimental group was lower than that of the control rats(P<0.05). The SIRT1 expression in the liver of the experimental group(P<0.01) and kidney(P<0.05). The expression level of the protein was higher than that of the control group. Conclusion SIRT1 protein is low in lung tissue of passive smoking rats and high in liver and kidney tissues.
