苦参碱调控TGF-β/Smad2信号通路逆转大鼠胰腺纤维化的作用研究

【摘要】 目的研究苦参碱对大鼠胰腺纤维化TGF-β/Smad2信号通路的影响作用,探讨苦参碱在胰腺纤维化中的作用究。方法构建大鼠胰腺纤维化模型,并利用HE染色判断模型是否成功;实验分为:假手术组,模型组和模型+苦参碱组,利用HE检测各组病理结果;免疫组化检测各组胰腺组织α-SMA、TGF-β1、I型胶原表达结果;荧光定量PCR和Western blot检测各组胰腺组织Smad2、TβR1、TβR II的表达结果。结果模型构建后大鼠身体状态良好,无死亡现象,并通过HE结果说明腺管周围小腺管增生,腺泡细胞线粒体肿胀,间质少量纤维堆积,无细胞浸润。上述结果表明模型构建成功。假手术组无明显纤维化,无进行性改变;苦参碱组可见少量腺泡细胞线粒体肿胀。与假手术组相比,模型组的a-SMA、TGF-β1、I型胶原表达明显升高,差异具有统计学意义(P<0.05)。与模型组相比,苦参碱组的a-SMA、TGF-β1、I型胶原表达明显下降,差异具有统计学意义(P<0.05)。与假手术组相比,模型组的Smad2、TβR1、TβR II型胶原表达明显升高,差异有统计学意义(P<0.05)。与模型组相比,苦参碱组的Smad2、TβR1、TβR II表达明显下降,差异有统计学意义(P<0.05)。结论苦参碱可以通过下调TGF-β1、Smad2的表达抑制胰腺星状细胞的活化,此可能为抑制纤维化形成与发展的相关机制,为苦参碱应用于临床给予可靠的理论依据。

【Abstract】 Objective To study the effect of Matrine on TGF-β/Smad2 signaling pathway in rat pancreatic fibrosis and explore the role of matrine in pancreatic fibrosis. Methods the rat model of pancreatic fibrosis was constructed and HE staining was used to determine whether the model was successful. The experiment is divided into: sham operation group,model group and model +Matrine group,using HE to detect the pathological results of each group,and the results of α-SMA,TGF-β1 and I collagen expression were detected by immunohistochemistry,and the expression of Smad2、TβR Ⅰ and TβR II of pancreas tissue in each group is detected by fluorescence quantitative PCR and Western blot. Results After the model construction,the rats were in good physical condition,without death,and the results of HE showed the hyperplasia of small glands around the gland,the swelling of the acinar cell mitochondria,the accumulation of a small amount of interstitial fibers and no cell infiltration. The above results show that the model is constructed successfully. In sham operation group,there was no obvious fibrosis and no progressive changes.In matrine group,a small amount of acinar cells mitochondria swelled. Compared with sham operation group,the expression of α-SMA,TGF-β1 and I collagen in the model group were significantly increased,and the difference was statistically significant(P<0.05). Compared with the model group,the expression of α-SMA,TGF-1 and type I collagen in matrine group decreased significantly,and the difference was statistically significant(P<0.05). Compared with the sham group,the expressions of Smad2,T R1 and T R type II collagen in the model group were significantly increased,and the difference was statistically significant(P<0.05). Compared with the model group,the expressions of Smad2,T R1 and T R II in matrine group were significantly decreased,and the difference was statistically significant(P<0.05). Conclusion Matrine can inhibit the activation of pancreatic stellate cells by reducing the expression of TGF-β1 and Smad2,which may be related to the mechanism of inhibiting the formation and development of fibrosis and provide a reliable theoretical basis for the application of matrine in clinical practice.
