绿茶茶多糖的抗氧化活性及对人肺癌细胞的抑制作用

【摘要】 目的探讨绿茶茶多糖的抗氧化活性及对人肺癌细胞A549的作用。方法采用热水浸提-乙醇沉淀法提取茶多糖,纯化后进行抗氧化活性检测,并采用MTT法检测不同浓度(0μg/mL、50μg/mL、100μg/mL、150μg/mL、200μg/mL)茶多糖对A549细胞增殖影响,采用流式细胞仪检测不同浓度茶多糖对A549细胞凋亡的影响,Western blot方法检测不同浓度茶多糖对Caspase-3、Bax及p53等蛋白表达影响。结果不同浓度绿茶茶多糖对DPPH自由基、超氧阴离子、羟基自由基及ABTS自由基均有一定清除作用,且清除作用呈剂量效应(P<0.05);不同浓度绿茶茶多糖均能抑制A549细胞增殖、促进凋亡,同时上调A549细胞中Caspase-3、Bax及p53蛋白表达,并呈剂量效应(P<0.05)。结论绿茶茶多糖抑制A549细胞增殖、促进凋亡与上调Caspase-3、Bax及p53蛋白表达相关。

【Abstract】 Objective To investigate the antioxidant activity and effect of polysaccharide in green tea on human lung cancer cells A549. Methods Tea polysaccharide was extracted by hot water extraction-ethanol precipitation method and purificated. The effects of tea polysaccharide at different concentrations(0 μg/mL, 50 μg/mL, 100 μg/mL, 150 μg/mL, 200 μg/mL) on proliferation of A549 cells were detected by MTT method, flow cytometry was used to detect the apoptosis of A549 cells. Western blot was used to detect the expression of Caspase-3, Bax and p53. Results Different concentrations of green tea polysaccharides have scavenging effects on DPPH radicals, superoxide anions, hydroxyl radicals and ABTS radicals with dose-dependent manner(P<0.05), inhibited the proliferation and promoted apoptosis of A549 cells, and up-regulated the expression of Caspase-3, Bax and p53 in A549 cells with dose-dependent manner. Conclusion Green tea polysaccharides could inhibit A549 cell proliferation and promote apoptosis, which is related to the up-regulation of Caspase-3, Bax and p53 protein expression.
