miR-194-3p调节TGF-β诱导的上皮-间充质转化抑制乳腺癌细胞侵袭

【摘要】 目的研究miRNA-194-3p在TGF-β1诱导的乳腺癌MDA-MB-231细胞上皮间质转化(EMT)发生中的作用,探讨miR-194-3p抑制乳腺癌侵袭的机制。方法体外培养人乳腺癌细胞系MDA-MB-231,采用TGF-β1加入人乳腺癌细胞系MDA-MB-231细胞培养液中制备细胞模型,RT-PCR检测miR-194-3p在TGF-β1诱导前后的表达变化。采用miRNA拟似物和无关序列分别转染MDA-MB231的方法获得细胞模型。Western blot检测EMT标记分子(E-cadherin、Vimentin)的表达。Trasnwell实验检测miRNA-194-3p拟似物、TGFβ1和共同作用对乳腺癌细胞侵袭能力的影响。结果 TGF-β1诱导乳腺癌细胞发生EMT,TGF-β组中E-cadherin表达下调,Vimentin表达上调;过表达miRNA-194-3p抑制了TGF-β对E-cadherin、Vimentin的作用。在侵袭迁移实验中,TGF-β促进细胞的侵袭,过表达miRNA-194-3p可抑制TGF-β的促进作用。结论 MiR-194-3p调节TGF-β诱导的上皮-间充质转化抑制乳腺癌侵袭。

【Abstract】 Objective To study the role of miRNA-194-3p in the occurrence of epithelial interstitial transformation(EMT) in TGF-β1 induced breast cancer mda-mb-231 cells, and explore the mechanism of miR-194-3p inhibiting the invasion of breast cancer. Methods The cell model was prepared by adding TGF-β1 to human breast cancer MDA-MB-231 cells, and RT-PCR was used to detect the expression of miR-194-3p before and after TGF-β1 induction. The expression of EMT marker molecules(E-cadherin, Vimentin) was detected by Western blot. The effect of miRNA-194-3p on the invasion ability of breast cancer cells was detected by Transwell experiment. Results TGF-β1 induced the occurrence of EMT in breast cancer MDA-MB-231 cells, the expression level of E-cadherin was decrased, but increased for Vimentin expression in TGF-β group. Overexpression of miRNA-194-3p inhibited the effect caused by TGF-β1 on E-cadherin and Vimentin. In Transwell experiments, TGF-β promoted cell invasion, overexpression of miRNA-194-3p inhibited EMT induced by TGF-β. Conclusion MiR-194-3p inhibits the invasion of breast cancer MDA-MB-231 cells is related to regulating the epithelial-mesenchymal transformation induced by TGF-β.
