血三七正丁醇部位促骨折愈合机制研究
Study on the Mechanism of Promoting Fracture Healing by N-butanol Fraction of D.Don var. Forb.
【摘要】 目的研究血三七正丁醇部位促骨折愈合的作用机制。方法采用CCK-8法测定正丁醇部位提取物对成骨细胞MC3T3-E1的增殖作用;FACS分析正丁醇部位对成骨细胞增殖周期的影响;碱性磷酸酶ELISA法检测血三七正丁醇部位对成骨细胞碱性磷酸酶活性的影响;ELISA法测定血三七正丁醇部位对前列腺素E2含量的影响。结果血三七正丁醇部位低剂量刺激成骨细胞增殖,高剂量抑制细胞增长,且其增殖作用表现为S-phase比例的增加;该部位有一定的促进成骨细胞分泌碱性磷酸酶作用,且依然呈现低剂量促进;10~100 ng/L的该部位能够明显降低成骨细胞中PGE2的含量。结论血三七正丁醇提取物具有较好的促进骨折愈合的作用。
【Abstract】 Objective To Study on the mechanism of promoting fracture healing from n-butanol fraction in D.Don var. Forb.(PAF). Methods The method of CCK-8 was used to measured the proliferation of osteoblast MC3 T3-E1 about n-butanol extracts in this plant; the effect of proliferation cycle of osteoblasts was evaluated by the method of Flow cytometry(FACS) analysis. The activity of Alkaline phosphatase(ALP) was evaluated by the kit of ELISA; the content of Prostaglandin E2(PGE2) was detected by the kit of ELISA. Results Low doses of the fraction of n-butanol in PAF stimulated proliferation of osteoblasts and inhibited cell growth at high doses,and its proliferation appeared as an increase in the proportion of S-phase. The n-butanol fraction of PAF promoted the secretion of alkaline phosphatase by osteoblasts,and it still exhibited low dose promotion and high dose inhibition. The sample of fraction n-butanol in PAF could significantly reduce PGE2 content in osteoblasts for the dosage of 10~100 ng/L. Conclusion The extracts from n-butanolin PAF has a good effect on promoting fracture healing.
