超声波辅助提取高效液相色谱法测定菠萝叶中小麦黄素的含量

【摘要】 研究超声波辅助提取菠萝叶中小麦黄素的最佳提取工艺和含量测定。利用单因素实验和正交实验,考察料液比、提取时间、提取温度、超声功率4个因素对提取工艺的的影响;采用高效液相色谱法对小麦黄素含量进行测定。菠萝叶中小麦黄素提取的最佳工艺为m(菠萝叶)∶V(乙酸乙酯)=1.0∶25(g·mL-1),提取时间90min,超声提取温度75℃,超声功率192W,小麦黄素的平均收率可达0.494%(n=3)。当小麦黄素进样量在0.006~0.12 mg·mL-1范围内与峰面积分值有良好的线性关系,r=0.99997,加样平均回收率为100.86%(n=5),相对标准偏差为3.24%。该方法简便有效,结果准确可靠。

【Abstract】 Studying the optimum methodology of ultrasonic extracting tricin from the leaves of Ananas comosus and determining the content of tricin.By orthogonal experiment and single factor experiment,we investigated the effect of 4 factors containing ratio of solid to liquid,extraction time,extraction temperature,ultrasonic power.After the extraction,the content of tricin was measured by HPLC.The best extraction technology was m(the leaves of Ananas comosus):V(ethyl acetate)=1.0∶25(g·mL-1),extraction time was 90 min,extraction temperature was 75℃,ultrasonic power was 192 W,theaverageyield of tricin was 0.494%(n=3),the amount of tricin had a excellent linear relationship with peak area when the amount of tricin ranged from 0.006~0.120 mg·mL-1,r=0.99997,average recovery rate was 100.86%(n=5),relative standard deviation was 3.24%.The method of extraction is feasible and effective,the results are accurate and reliable.
