二元表法和MCID法快速鉴定烟草品种
Rapid Identification of Tobacco Varieties by Using Binary Table Method and MCID Method
【摘要】 为建立快速鉴定烟草品种的方法,利用RAPD分子标记方法对13个烟草品种进行鉴定,分别应用二元表法和人工绘制品种树图(Manual cultivar identification diagram,MCID)法2种分析方法对烟草品种DNA指纹图进行分析。结果表明:从150条特异性引物中共筛选出4条稳定性好、特异性高的随机引物,得到4张DNA指纹图,利用2种分析方法对指纹图分析后成功得到13个品种的分子ID,并构建出13个烟草品种的MCID。研究发现,2种分析方法均可有效鉴定烟草品种,其中,MCID法的直观性和可加性优于二元表法,操作性更强,当供试品种较多时MCID法更简单实用,可用于大量烟草品种的快速鉴定。
【Abstract】 In order to establish a rapid identification method for tobacco varieties,this study used RAPD molecular marker to identify 13 tobacco varieties.Four random primers with good stability and high specificity were successfully screened from 150 random primers.DNA fingerprints were analyzed by binary table and manual cultivar identification diagram(MCID) methods,by which molecular IDs and the MCID of 13 tobacco varieties were successfully established.Although both molecular IDs and the MCID could be used to discriminate 13 tobacco varieties,the MCID method is more intuitive,more additive and more operational than binary table method.Therefore,the MCID method is better used for rapid identification of a large number of tobacco varieties.
