不同强度运动对糖尿病造模大鼠的血清IL-6、β2-MG及肾脏TGF-β1蛋白表达的影响

【摘要】 目的探究不同强度运动干预在糖尿病造模过程对血清白细胞介素6(interleukin-6,IL-6)、β2微球蛋白(β2 microglobulin,β2-MG)和肾脏转化生长因子β1(transforming growth factor-β1,TGF-β1)的影响。方法选取50只SD雄性大鼠,随机分为正常对照组、糖尿病对照组、低强度运动组、中等强度运动组和高强度运动组5组,每组10只。正常对照组普通饲料喂养,糖尿病对照组、低强度运动组、中等强度运动组、高强度运动组高糖高脂喂养,糖尿病对照组笼中自由活动,低强度运动组、中等强度运动组、高强度运动组实验期间分别进行不同负荷的跑台运动。肾脏组织以过碘酸六胺银(periodic acid-silver methe-namine,PASM)染色。酶联免疫吸附测定(enzyme-linked immunosorbent assay, ELISA)检测大鼠血清中IL-6、全自动生化仪检测血清β2-MG的浓度。Western blot检测大鼠肾脏组织TGF-β1蛋白表达。结果 PASM染色结果显示正常对照组肾小球结构清晰,基底膜无明显改变;糖尿病对照组、低强度运动组肾小球基底膜有所增厚,系膜区明显增宽;中等强度运动组、高强度运动组肾小球基底无明显增厚,系膜区轻微增宽。ELISA结果显示,与正常对照组、糖尿病对照组相比,低强度运动组和中等强度运动组血清IL-6水平均明显下降(P<0.05)。各组大鼠之间血清β2-MG水平差异无统计学意义(P>0.05)。Western blot结果表明各组间TGF-β1的表达水平差异无统计学意义(P>0.05)。结论在糖尿病造模过程中,8周耐力运动可通过降低机体血清IL-6水平并保护肾脏组织结构,且中等强度耐力运动保护作用最为显著。

【Abstract】 Objective To explore the effects of different intensity exercises interventions on serum interleukin-6(IL-6), β2 microglobulin(β2-MG) and renal transforming growth factor-β1(TGF-β1) during diabetes modeling. Methods Fifty SD male rats were randomly divided into normal control group, diabetic control group, low-intensity exercise group, moderate-intensity exercise group and high-intensity exercise group,each group with 10 rats. The rats in normal control group were fed with ordinary fodder, the rats in diabetic control group were fed with high-sugar and high-fat diet and with free movement in cage. The rats in low-intensity exercise group, moderate-intensity exercise group and high-intensity exercise group were fed with high-sugar and high-fat diet and along with different levels of treadmill exercise during the experiment,respectively. The kidney tissue were observed by periodic acid-silver methenamine(PASM) staining, the expressions of serum IL-6 were detected by enzyme-linked immunosorbent assay(ELISA),the concentration of serum β2-MG were detected by automatic biochemical analyzer and the protein expressions of TGF-β1 in rats’ kidney tissue were detected by Western blot.Results The results of PASM showed that the glomerular structure was cleared and the basement membrane was not changed obviously in normal control group. The glomerular basement membrane was thickened and the mesangial area was widened obviously in low-intensity exercise group and diabetic control group. The glomerular base was not significantly thickened and the mesangial area was widened slightly in moderate-intensity exercise group and high-intensity exercise group. The serum IL-6 levels in low-intensity exercise group and moderate-intensity exercise group were significantly lower than those in normal control group and diabetic control group(P<0.05). There were no significant differences in the level of serum β2-MG and the expression of TGF-β1 among the groups(P>0.05).Conclusion During the modeling of diabetes, 8-week endurance exercise can reduce the level of serum IL-6 and protect the kidney tissue structure, and the moderate-intensity endurance exercise protection is the most significant.
