Prx Ⅱ缺失的真皮间充质干细胞提前衰老加速皮肤老化进程研究
Study of Peroxiredoxin Ⅱ Skin Aging Delay by Protecting Dermal Mesenchymal Stem Cells
【摘要】 通过比较野生型和Prx II(Peroxiredoxin II)基因敲除型小鼠皮肤老化情况,阐明皮肤老化过程中Prx II对真皮间充质干细胞的保护作用。体内试验采用病理组织切片,蛋白免疫印迹等方法检测两种129/SvJ小鼠皮肤组织及生理功能的差异;体外试验采用SA-β-gal染色对比两种真皮间充质干细胞衰老情况。结果表明同月龄的Prx II基因敲除型小鼠与野生型小鼠相比真皮厚度层变薄,伤口愈合速度减慢,皮肤组织中P16、p21表达水平增加,PCNA表达水平降低;同代数的Prx II基因敲除型的真皮间充质干细胞与野生型相比衰老细胞数量明显增多。综合分析,Prx II基因敲除小鼠皮肤细胞周期蛋白依赖性激酶抑制因子表达上升,皮肤发生提前老化,与真皮间充质干细胞的细胞衰老存在相关性,为通过Prx II延缓皮肤老化,减少因皮肤老化而引发的多种疾病提供基础理论。
【Abstract】 To explore the protective effect of Prx II on dermal mesenchymal stem cells(DMSCs)during the skin aging progress,the skin aging of Prx II gene knockout mouse(Homo)was compared with that of wild type mouse(WT). In vivo,the difference of 129/SvJ skin tissues and physiological function of two kinds of mouse was tested by Histopathologic slide and Western blotting;in vitro,aging of two DMSCs was compared using SA-β-gal staining. The results showed that the dermal thickness of Homo was thinner than that of WT and healing was slower at the same age. the expression levels of P16 and P21 were increased while that of PCNA were decreased.Meanwhile,the number of senescent cells was significantly increased in Homo. In summary,the expression of protein-dependent kinase inhibitors increased in Homo,the early skin aging was relative to the aging of DMSCs in mouse,which provided fundamental basis for reducing kinds of illness caused by skin aging through the function of delay the skin aging by Prx II.
