人乳腺癌细胞中热休克蛋白90的表达及与细胞增殖、凋亡的关系
The expression of apoptosis HSP90 genes in breast cancer and its relation with cell apoptosis and proliferation
【摘要】 目的探讨热休克蛋白(HSP) 90在乳腺癌细胞中的表达及与乳腺癌细胞凋亡及增殖的相关性。方法应用免疫组化聚合酶两步法(PV法)检测乳腺正常组织、乳腺腺瘤及乳腺癌中HSP90的表达情况,Ki67检测细胞增殖情况,TUNEL试剂盒检测细胞凋亡情况。结果 HSP90的表达与乳腺癌细胞凋亡指数(AI)呈负相关,而与增殖指数(PI)呈正相关(P <0. 05)。结论乳腺癌的发生、发展中可能普遍存在细胞凋亡、增值机制的紊乱,HSP90的异常表达在乳腺癌的发生、发展中起重要作用。
【Abstract】 Objective To investigate the expression of apoptosis HSP90 genes in breast cancer and its relation with apoptosis and proliferation of breast cancer cells. Methods Immunohistochemical method( PV) was used to detect the expression of HSP90 in normal mammary gland tissues,mammary gland adenoma and breast cancer,and the cell proliferation was measured by means of Ki67,moreover,TUNEL kit was used to detect the cell apoptosis. In addition the confocal microscopy was used to evaluate the apoptosis of tumor cells. Results HSP90 expression was negatively related to AI,however,which was positively related to PI( P < 0. 05). Conclusion There exists the disturbance of cell apoptosis and proliferation during the pathogenesis and development of breast cancer. The abnormal expressions of HSP90 may play an important role in the pathogenesis and development of breast cancer.
