丹参提取物对肿瘤合并心肌缺血损伤的保护效应
Protective effect of Salvia miltiorrhiza extract on tumor with myocardial ischemic injury
【摘要】 目的观察丹参提取物在肿瘤合并心肌缺血模型中的作用。方法 30只雌性BALB/C-nu 4~6周龄裸鼠皮下接种乳腺癌MCF-7细胞,制备移植瘤,选取成功移植瘤的18只小鼠随机分为3组,即肿瘤模型组、药物丹参酮ⅡA组、药物丹酚酸B组,每组6只。移植瘤第7天后,每组给予腹腔注射异丙肾上腺素2.5 mg/kg,连续注射7 d,制备肿瘤合并心肌缺血动物模型。另设正常对照组6只。取出肿瘤组织称重,心脏组织进行Masson′s Trichrome染色。采用免疫组织化学方法检测各组心肌和肿瘤组织中CD34和血管性血友病因子(von Willebrand Factor, vWF)的表达,采用MTT方法观察丹参酮ⅡA对人脐静脉血管内皮细胞(human umbilical vein endothelial cells,HUVECs)及乳腺癌MCF-7增殖活力的影响。结果肿瘤模型组心肌胶原纤维面积比较正常对照组明显增多;丹参酮ⅡA组及丹酚酸B组肿瘤重量和心肌胶原纤维面积比明显小于肿瘤模型组(P<0.05);在心肌组织中,丹参酮ⅡA组和丹酚酸B组CD34和vWF蛋白表达水平较肿瘤模型组明显升高(P<0.05);在肿瘤组织中,丹参酮ⅡA组和丹酚酸B组CD34和vWF蛋白表达水平较肿瘤模型组明显降低(P<0.05)。体外实验结果显示,丹参酮ⅡA能够促进HUVEc增殖,抑制乳腺癌细胞增殖,均具有剂量依赖性。结论丹参酮ⅡA有保护心肌缺血和抑制乳腺癌细胞的生长的作用,可能与血管生成有关。
【Abstract】 Objective To observe the role of Salvia miltiorrhiza extract in tumor with myocardial ischemia model. Methods Thirty female BALB/C-nu nude mice which were 4-6 week old were subcutaneously inoculated with breast cancer MCF-7 cells to prepare transplanted tumors. Eighteen mice with successful transplanted tumors were randomly divided into three groups: tumor model group,tanshinone ⅡA group and salvianolic acid B group. After 7 days of transplantation, each group was injected with isoproterenol 2.5 mg/kg intraperitoneally for 7 days to establish an animal model of tumor with myocardial ischemia. Another 6 rats were set as normal control group. The tumor tissue was removed and weighed, and the heart tissue was subjected to Masson′s Trichrome staining. The expression of CD34 and von Willebrand Factor(vWF) in myocardium and tumor tissues was detected by immunohistochemistry. The effect of tanshinone ⅡA on human umbilical vein endothelial cells(HUVECs) and breast cancer MCF-7 were observed by MTT. Results The results showed that the area of myocardial collagen fibers in the tumor model group was significantly higher than that in the normal control group. The tumor weight and myocardial collagen fiber area in the tanshinone ⅡA group and salvianolic acid B group was significantly smaller than that in the the tumor model group(P<0.05). In the myocardial tissue, the expression levels of CD34 and vWF protein in the tanshinone ⅡA group and the salvianolic acid B group were significantly higher than those in the tumor model group(P<0.05). In the tumor tissue, the expression levels of CD34 and vWF protein in tanshinone ⅡA group and in salvianolic acid B were significantly lower than those in the tumor model group(P<0.05). In vitro experiments showed that tanshinone ⅡA can promote the proliferation of HUVECc and inhibit the proliferation of breast cancer cells in a dose-dependent manner. Conclusion The effect of tanshinone ⅡA on protecting myocardial ischemia and inhibiting the growth of breast cancer cells may be related to angiogenesis.
