小麦光敏色素互作因子TaPIF4基因的克隆与表达分析
Cloning and Expression Analysis of Phytochrome Interacting Factor TaPIF4 in Wheat
【摘要】 为研究小麦光敏色素互作因子TaPIF4基因的结构、特性以及在小麦中的具体功能,从小麦中克隆TaPIF4基因并进行生物信息学分析,采用实时荧光定量PCR(qRT-PCR)分析该基因在48 h内的节律表达及在低温、干旱、高盐和脱落酸(ABA)处理条件下的表达模式,推测其可能参与的生物学过程。结果表明,TaPIF4基因的开放阅读框为1 053 bp,编码350个氨基酸,所编码的蛋白在C端含有1个b HLH结构域。TaPIF4蛋白的b HLH结构域保守程度高,在进化关系上与粗山羊草的PIF4-like蛋白最近。TaPIF4基因的表达具有昼夜节律性,在光照条件下表达量低,在黑暗条件下表达量高。TaPIF4基因在外源ABA处理下表达被明显抑制;在干旱处理下,短时间内其表达量升高,之后下降;冷处理下,表达模式与干旱处理相反;盐处理下,其表达无明显规律。推测TaPIF4基因可能参与植物ABA、干旱、低温胁迫的信号通路。
【Abstract】 In order to study the structure,characteristics and function of phytochrome interacting factor TaPIF4 gene in wheat,the TaPIF4 gene was cloned from wheat and analyzed by bioinformatics methods. We analyzed the rhythmic expression of this gene within 48 hours and its expression pattern under the stress conditions of low temperature,drought,high salt and abscisic acid( ABA) and speculated the possible biological processes by Real time fluorescence quantitative PCR( qRT-PCR). The results showed that the open reading frame( ORF) of TaPIF4 gene was 1 053 bp,and 350 amino acids were encoded. The protein encoded by this gene contained a bHLH conserved domain at the C-terminus. The bHLH domain of TaPIF4 protein was highly conserved and TaPIF4 protein was closest to the PIF4-like protein of Aegilops tauschii in evolution. The expression of TaPIF4 gene had circadian rhythm,which was characterized by low expression under light conditions and high expression under dark conditions. The expression of TaPIF4 gene was obviously inhibited by exogenous ABA. Under drought conditions,the expression of TaPIF4 increased in early stage and declined in later stage. At low temperature,the expression pattern of TaPIF4 was opposite to that of drought conditions. Under the salt stress,there was no apparent regular pattern in the expression of TaPIF4. It is speculated that TaPIF4 may be involved in plant ABA,drought and low temperature stress signal pathway.
