【摘要】 目的建立HPLC法测定ATP竞争性mTOR抑制剂INK128的含量。方法采用Lichrospher CN（250mm×4.6mm,5μm）色谱柱;以乙腈为流动相A,以0.01mol·mL^-1磷酸二氢铵为流动相B,进行梯度洗脱（0～40min,5%A→15%A;40～50min,15%A→5%A）,流速1.0mL·min^-1,检测波长247nm;柱温30℃,进样量20μL。结果 INK128在50.16～802.56μg·mL^-1范围内与色谱峰面积呈良好线性关系;精密度RSD为0.29%;平均回收率（n=9）为99.6%。结论本方法准确可靠,重复性好,可用于INK128的质量控制。更多还原

【Abstract】 OBJECTIVE To establish a HPLC method to determine ATP-competitive mTOR inhibitor INK128.METHODS The analysis was performed on a Lichrospher CN column（250 mm×4.6 mm,5μm）with gradient mobile phase of acetonitrile-0.01 mol·mL^-1 ammonium dihydrogen phosphate solution at a flow rate of 1.0 mL·min^-1.The detection wavelength was set at 247 nm,and the column temperature was 30℃,the injection volume was 20μL.RESUITS The linear range of INK128 was 50.16～802.56μg·mL^-1,the precision RSD was 0.29%,and the average recoveries（n=9） was 99.6%.CONCLUSION The method is accurate and good repeatability,can be used for quality control of INK128.更多还原
