多梳基因蛋白EZH2促进喉鳞状细胞癌血管生成的机制及临床评价
Mechanism and clinical evaluation of polycomb group protein EZH2 in promoting angiogenesis in laryngeal squamous cell carcinoma
【摘要】 目的检测果蝇zeste基因增强子同源物2(Enhancer of zeste homolog 2,EZH2)和血小板-内皮细胞黏附分子31(platelet endothelial cell adhesion molecule-31,CD31)在喉鳞状细胞癌(laryngeal squamous cell carcinoma,LSCC)、喉正常黏膜(normal laryngeal tissues,NLT)中的表达,并探讨EZH2促进喉鳞癌血管生成的机制及EZH2在LSCC发生发展中的作用。方法收集LSCC标本61例、NLT标本20例,采用免疫组化SP法检测该两组标本中CD31、EZH2的表达。结果①LSCC中以CD31标记的微血管密度(microvessel density,MVD)为10.26±2.46,大于NLT(1.20±1.10,t=-15.85,P<0.05)。CD31标记的MVD(CD31-MVD)的表达与LSCC的T分级、临床分期、淋巴结转移、是否复发有关(P均<0.05)。②EZH2蛋白表达的阳性率在LSCC为65.57%(40/61),大于NLT 30.00%(6/20)(χ~2=23.92,P<0.05)。EZH2蛋白表达与LSCC的组织学分级、T分级、临床分期、淋巴结转移、是否复发有关(P均<0.05)。③CD31和EZH2蛋白表达呈正相关。④生存分析显示,CD31、EZH2、淋巴结转移、临床分期、有无复发是患者预后独立的影响因素(P<0.05)。结论 EZH2蛋白在LSCC中表达上调,促进LSCC新生血管生成、发展、浸润、转移及预后较差,为LSCC的治疗提供新的分子靶点和理论依据。
【Abstract】 Objective To detect the expression of enhancer of zeste gene enhancer homolog 2(EZH2) and platelet endothelial cell adhesion molecule 31(CD31) in laryngeal squamous cell carcinoma(LSCC) and normal laryngeal tissues(NLT) of drosophila melanogaster, and to explore the role of EZH2 in promoting blood flow of laryngeal squamous cell carcinoma. The mechanism of tube formation and the role of EZH2 in the occurrence and development of LSCC were discussed. Methods 61 LSCC specimens and 20 NLT specimens were collected. The expressions of CD31 and EZH2 in the two groups were detected by immunohistochemical SP method. Results ①The mean of Microvessel density(MVD) value marked by CD31 in LSCC was10.26±2.46, which was higher than that in NLT(1.20±1.10,t=-15.85,P<0.05). The expression of MVD marked by CD31(CD31-MVD) was correlated with T stage, clinical stage, lymph node metastasis and recurrence in LSCC(P<0.05). ②The positive expression rate of EZH2 protein in LSCC was 65.57%(40/61), which was higher than that in NLT 30.00%(6/20)(χ~2=23.92,P<0.05). The expression of EZH2 was correlated with histological grading, T stage,clinical stage, lymph node metastasis, and recurrence in LSCC(P<0.05). ③There was a positive correlation between CD31 and EZH2 protein.④Survival analysis indicated that the expressions of CD31 and EZH2, lymph node metastasis,clinical stage,and recurrence were independent factors for tumor prognosis in LSCC(P<0.05). Conclusion The up-regulation of EZH2 protein in LSCC promotes angiogenesis, development, invasion, metastasis and poor prognosis of LSCC, and provides a new molecular target and theoretical basis for the treatment of LSCC.
