一测多评法测定玫瑰精油中14种有效成分的含量
Analysis of fourteen effective components in rose essential oil by quantitative analysis of multi-components by single-marker
【摘要】 目的:采用一测多评法测定玫瑰精油中柠檬烯、正戊醇、玫瑰醚、正十五烷、芳樟醇、正十七烷、香茅醇、橙花醇、乙酸苯乙酯、香叶醇、苯乙醇、正二十烷、正二十一烷、正二十三烷含量,同时采用内标法测定3批不同精油中14种化学成分的含量,比较计算值与实测值的差异,验证一测多评法的准确性。方法:采用气相色谱法,DB-WAX毛细管色谱柱(30 m×0. 32 mm×0. 25μm),氢火焰离子化检测器(FID),程序升温,检测器温度为250℃,进样口温度为240℃,分流比为35∶1,以正丁醇为内标物,以正十七烷为参照物,确定13种化学成分之间的相对校正因子,并进行含量计算,实现一测多评。结果:仪器精密度、重复性和稳定性良好,各化合物在线性范围内线性关系良好(R2≥0. 9985),14种化合物平均回收率介于97. 04%~104. 88%(RSD<4%,n=9),内标法与一测多评法测定三批样品没有显著性差异(RSD<3%)。结论:本研究建立的一测多评方法操作简单,测定结果准确,能较大程度节约成本,作为一种新模式用于玫瑰精油中多成分的定量分析,也可为全面监控玫瑰精油质量提供参考。
【Abstract】 In this study,quantitative analysis of multi-components by single marker was adopted to analyze the content of 14 components( limonene,1-pentanol,rose oxide,n-pentadecane,linalool,n-heptadecane,citronellol,nerol,phenylethyl acetate,geraniol,phenethyl alcohol,n-eicosane,n-heneicosane,and n-tricosane) in rose essential oil. At the same time,the contents of the 14 components in 3 batches of rose essential oil were determined by internal standard method. Through comparison of the difference between the calculated value and the measured value,the accuracy of the QAMS method was verified. The parameters for gas chromatography( GC) were as follows: Agilent’s DB-WAX capillary column( 30 m × 0. 32 mm × 0. 25 μm) was used with FID,detector temperature was250℃,inlet temperature was 240℃,split ratio was 35 ∶ 1. Then n-butanol was used as the internal standard,and n-heptadecane was used as the reference substance to determine the relative correction factors( RCF) of the 13 components,and the contents of the components were calculated,thus realizing quantitative analysis of multi-components by single-marker. The results showed that the instruments’ precision,repeatability and stability were good,the linear relationship of the components in the linear range was good( R2≥0. 9985),the average recovery of the 14 compounds ranged from 97. 04% to 104. 88%( RSD<4%,n = 9),and there was no significant difference between the internal standard method and the QAMS method( RSD<3%). The method was simple,accurate and cost-effective,which could be used for quantitative analysis of multi-components in rose essential oil. This study also provided reference for comprehensive monitoring of the quality of rose essential oil.
