N,N’-二环己基-N-亚麻酸酰脲的体外抗肿瘤活性
Anti-tumor activity of N,N’-dicyclohexyl-N-linolenic acid urea in vitro
【摘要】 目的探讨板蓝根组酸衍生物N,N’-二环己基-N-亚麻酸酰脲(化合物MSR405)的体外抗肿瘤活性。方法取对数生长期肝癌细胞Bel-7402、Hep-G2、SMMC-7721和卵巢癌细胞SKOV3,均分别应用500μg/mL、250μg/mL、125μg/mL、62.5μg/mL、31.25μg/mL MSR405进行干预,检测细胞的增殖抑制情况。应用60μg/mL、80μg/mL、100μg/mL、120μg/mL MRS405干预肝癌细胞Bel-7402、SMMC-7721,以未进行干预的细胞作为阴性对照,24 h后检测细胞的凋亡情况,并观察细胞形态。结果不同浓度MSR405作用下,Bel-7402、SMMC-7721、Hep-G2和SKOV3细胞的增殖受到不同程度地抑制。与阴性对照组比较,经各浓度MSR405处理后肝癌细胞Bel-7402、SMMC-7721的凋亡率均升高(均P <0.05); MSR405浓度为80~120μg/mL时,Bel-7402和SMMC-7721细胞的凋亡率随着MSR405浓度增大而升高(均P <0.05)。各药物组的细胞多呈凋亡细胞的形态。结论不同浓度MSR405对肝癌及卵巢癌细胞的增殖均有不同程度的抑制作用,在一定浓度范围内其对肝癌细胞Bel-7402、SMMC-7721的促凋亡作用具有浓度依赖性。
【Abstract】 Objective To explore in vitro anti-tumor activity of Radix isatidis acid derivative N,N’-dicyclohexyl-N-linolenic acid urea( compound MSR405).Methods Intervention with 500 μg/mL,250 μg/mL,125 μg/mL,62.5 μg/mL,and 31.25 μg/mL MSR405 was performed,respectively,in hepatoma cells Bel-7402,Hep-G2,SMMC-7721 and ovarian cancer cell SKOV3 in logarithmic phase,to detect their inhibition on proliferation of the cells.Intervention with 60 μg/mL,80 μg/mL,100 μg/mL,and 120 μg/mL MRS405 was conducted,respectively,in hepatoma cells Bel-7402 and SMMC-7721,with cells without intervention as negative controls,apoptosis of the cells was determined 24 hours later,and cellular morphology was observed.Results Different concentrations of MSR405 exerted inhibitions on the proliferation of Bel-7402,SMMC-7721,Hep-G2 and SKOV3 cells in different degrees.Compared with the negative control group,hepatoma cells Bel-7402 and SMMC-7721 treated with various concentrations of MSR405 exhibited increased apoptosis rates( all P < 0.05); when the concentration of MSR405 ranged from 80 to 120 μg/mL,the apoptosis rates of Bel-7402 and SMMC-7721 cells increased with the increasing MSR405 concentration( all P < 0.05).Cells in the medication groups commonly presented their structure as apoptotic cells’.Conclusion Different concentrations of MSR405 have inhibitory effects on the proliferation of hepatoma and ovarian cancer cells in different degrees,which,within a certain concentration limits,might exert a pro-apoptotic effect on hepatoma cells Bel-7402 and SMMC-7721 in a dose-dependent manner.
