标本离体固定时间对荧光原位杂交技术检测乳腺浸润性导管癌HER2基因的影响
Impact of in vitro specimen fixation time on HER2 gene detected by fluorescence in situ hybridization in breast invasive ductal carcinoma
【摘要】 目的探讨标本离体固定时间对荧光原位杂交(FISH)技术检测乳腺浸润性导管癌HER2基因的影响。方法选择原发性乳腺浸润性导管癌患者116例,将每例患者同一癌组织标本分成5份,根据标本离体固定时间将标本分为A组(5 min)、B组(30 min)、C组(60 min)、D组(90 min)、E组(120 min),每组各116例。采用FISH法检测各组HER2基因,观察各组HER2基因荧光镜检结果,比较各组HER2基因阳性检出率。结果 A、B、C组荧光显微镜下的细胞轮廓完整性、背景干净度均优于D、E组,且橘红/绿色荧光信号强度及数量均强于或多于D、E组。A、B、C组HER2基因阳性检出率均高于D、E组(均P <0. 005)。结论标本离体固定时间可影响FISH法检测乳腺浸润性导管癌HER2基因的结果,标本离体固定时间以30 min内为宜,最长不应超过60 min。
【Abstract】 Objective To investigate the impact of in vitro specimen fixation time on HER2 gene detected by fluorescence in situ hybridization( FISH) in breast invasive ductal carcinoma. Methods A total of 116 patients with primary breast invasive ductal carcinoma were enrolled,and cancer tissue specimen from each patient was divided into five copies,according to in vitro specimen fixation time,the specimens were divided into group A( 5 minutes),group B( 30 minutes),group C( 60 minutes),group D( 90 minutes) and group E( 120 minutes),with 116 cases in each group. HER2 gene was detected by FISH assay in each group,fluorescence microscopy results of HER2 gene in each group were observed,and the positive rate of HER2 gene was compared among groups. Results Compared with group D or E,groups A,B and C showed superior integrity of cell contour and background cleanliness under fluorescence microscope,along with orange/green fluorescent signals stronger in intensity and more in quantity. The positive rate of HER2 gene in group A,B or C was higher than that in group D or E( all P < 0. 05). Conclusion In vitro specimen fixation time may influence the results of FISH assay for detecting HER2 gene in breast invasive ductal carcinoma,the appropriate in vitro specimen fixation time is defined as less than 30 minutes,with the longest time no more than60 minutes.
