【摘要】 目的:探讨广西红水河流域长寿人群生长激素促分泌受体(GHSR)rs572169基因多态性对血糖和血脂的影响,以了解该位点多态性对该地区长寿所起的可能作用。方法:选取广西红水河流域长寿带壮族居民1 830例,分为长寿组(年龄≥90岁长寿老人,n=496)、子女组(长寿老人的第一代子女,n=723)及对照组(追溯3代以上均无年龄≥90岁老人,n=611)。应用多重高温连接酶检测反应技术(iMLDR)对GHSR基因进行分型,分析该基因位点多态性对空腹血糖(FBG)及血脂指标影响。结果:长寿组FBG水平低于子女组和对照组(P<0.05);长寿组甘油三酯(TG)水平较其他两组低,且高密度脂蛋白胆固醇(HDLC)水平高于对照组(均P<0.05)。女性人群中,对照组CT/TT基因型及T等位基因明显高于长寿组(P<0.05)。在对照组中,CT/TT基因型总胆固醇(TC)水平高于CC型基因型人群(P<0.05)。进行性别分层后发现,在总体和子女组人群中,女性群体的TG水平以及对照组男性群体的血糖水平,均为CT/TT基因型高于CC基因型人群(P<0.05)。结论:GHSR rs572169T多态性与长寿人群的血糖和血脂有一定的关系,但受性别因素的影响。

【Abstract】 Objective:To investigate the effect of growth hormone secretagogue receptor(GHSR) rs572169 polymorphism on blood lipid and blood glucose levels in the longevity population in Guangxi Hongshui River Basin.Methods:Individuals were enrolled from the longevity region Hongshui River Basin in Guangxi of China,and were divided into longevity group(age≥90 years old,n=496),offspring group(the first generation of the longevity group,n=723) and control group(no family members over 90 year old in 3 generations,n=611).The improved multiple ligase detection reaction(iMLDR) technique was employed to genotype SNP rs572169.Results:The fasting blood glucose(FBG) level of longevity group was lower than that of other two groups(P<0.05).Longevity group exhibited a lower triglyceride(TG) level and a higher level of density lipoprotein cholesterol(HDLC) as compared to the control group(P<0.05).Females in longevity group had a lower T allele genotype(CT/TT) frequency than the controls(P<0.05).In the control group,the total cholesterol(TC) level of subjects with CT/TT genotype was higher than those with CC genotype.Female CT/TT carriers in offspring group and total population presented higher TG level,while male CT/TT carriers in control group presented higher FBG level compared with CC carriers(P<0.05).Conclusion:GHSR rs572169 T polymorphism was associated with blood glucose and blood lipids in longevity population,but it was influenced by gender factor.
