同种异基因心脏移植术后移植物浸润白细胞的动态变化研究
Study on graft infiltrating cells proportion changes after allogenic heart transplantation
【摘要】 目的:分析同种异基因心脏移植术后移植物内浸润白细胞的动态变化。方法:采用Balb/c和C57小鼠分别作为供体和受体,建立颈部心脏移植模型,分别在移植后第1、第3、第5、第7天处死小鼠,获取心脏移植物,使用流式细胞仪检测移植物浸润白细胞的比例;苏木精—伊红(HE)染色,观察移植术后心脏的病理改变。结果:心脏移植物内浸润白细胞中,术后第1天主要为CD11b~+Ly-6G~+中性粒细胞和CD11b~+F4/80巨噬细胞,比例分别为(32±2.6)%和(16.6±1.1)%,术后第3天,主要为CD11b~+F4/80巨噬细胞、NK1.1NK细胞和CD3~+CD4~+T辅助细胞,分别占(49.6±4.5)%、(17.6±2.9)%和(15.2±1.1)%,术后第5、第7天主要为CD11b~+F4/80巨噬细胞和CD3~+CD4~+T辅助细胞。病理学检测发现,移植后第1天心脏移植物内见白细胞浸润,心肌病理结构基本正常;但是移植术后第3天,白细胞浸润明显增多,心肌间质出血、心肌水肿;术后第5天和第7天,白细胞浸润继续增加,心肌正常结构严重破坏。结论:器官移植术后急性排斥反应中先天性固有免疫反应占有重要作用。
【Abstract】 Objective:To investigate graft infiltrating cells proportion changes early allogenic heart transplantation through flow cytometry analysis.Methods:Balb/c mice and C57 mice were used as donor and recipient respectively to establish cervical heart transplant model.The recipient mice were sacrificed on day 1,3,5,and 7 after transplantation,and the heart grafts were harvested to analyze graft infiltrating cells proportion through flow cytometry.Histological changes were evaluated by HE staining.Results:On day 1,the graft infiltrating cells were mainly CD11 b+Ly-6 G+ neutrophils and CD11 b+F4/80 macrophages,the proportion were 32%±2.6% and 16.6%±1.1%,respectively.They were mainly CD11 b+F4/80 macrophages,NK1.1 NK cells and CD3+CD4+ Th cells in the graft on day 3 and the cells propotion were 49.6%±4.5%,17.6%±2.9% and 15.2%±1.1%.On day 5 and 7,the graft infiltrating cells were mainly CD11 b+F4/80 macrophages and CD3+CD4+Th cells.Pathological examinations found relatively rare graft infiltrating cells and normal heart muscle structure on day 1.However,the grafting infiltrating cells increased on day 3 and myocardial interstitial hemorrhage,myocardial edema were observed.On day 5 and day7,more severe graft infiltrating cells and damaged heart muscle were observed.Conclusion:Innate immune response play key role in acute immune rejection after organ transplantation.
