二维高效液相色谱法测定人血清中部分抗精神病药物浓度体系的建立

【摘要】 目的建立一套快速、准确的二维高效液相色谱法(2D-LC-UV),用于临床上测定人血清中氯氮平、奥氮平、利培酮、帕利哌酮、喹硫平、阿立哌唑及齐拉西酮7种常用的第二代抗精神病药物的血药浓度。方法联用二维液相色谱耦合仪和高效液相色谱仪并建立相应的方法学及标准曲线,血清样品经过前处理后通过自动进样器进行分析。结果空白血清对药物的检测无干扰,氯氮平、奥氮平、利培酮、帕利哌酮、喹硫平、阿立哌唑及齐拉西酮各药物特征峰出峰良好,并且在其方法学的线性范围内线性良好,相关系数r> 0.9996;日内、日间精密度(RSD%)均小于10%,回收率85%~115%。结论用二维高效液相色谱法(2D-LC-UV)这一体系进行抗精神病药物浓度测定,前处理操作简单,能够快速、准确地检测所选择的7种抗精神病药物的血药浓度。

【Abstract】 Objective To establish a rapid and accurate two-dimensional high performance liquid chromatography(2D-LC-UV),using in clinical determination of clozapine,olanzapine,risperidone,paliperidone,quetiapine,aripiprazole and ziprasidone in human serum. Methods Two-dimensional liquid chromatography coupled with high performance liquid chromatography was used to establish the corresponding methodology and standard curve. Serum samples were analyzed by automatic sampler after pre-treatment. Results The blank serum did not interfere in the drug detecting assay. The characteristic peaks of clozapine,olanzapine,risperidone,paliperidone,quetiapine,aripiprazole and ziprasidone were well formed,as well as the linearity in the linear range of its methodology,with a correlation coefficient r > 0.9996. The intra-day and inter-day precision(RSD%)were less than 10%. The recoveries ranged from 85% to 115%. Conclusion The pretreatment operation is simple with two-dimensional high performance liquid chromatography(2D-LC-UV)in determining the concentration of antipsychotics,with which the serum concentration of the seven selected antipsychotics can be rapidly and accurately detected.
