西藏野生韭遗传多样性的初步研究
The preliminary study on genetic diversity of Allium hookeri Thwaitesfrom the Tibet
【摘要】 西藏野生韭广泛分布于西藏,具有特殊气味,受到广大西藏人民的喜爱。但是遗传背景鲜有研究。本研究利用SRAP技术,利用了10对引物对11份西藏野生韭进行SRAP分析发现:10对引物组合共扩增出835条谱带,其中多态性条带共826条,多态性比例为96.83%。11个样品之间的遗传相似系数在0.355-0.575之间,平均相似系数为0.463,说明不同海拔高度采集的四个种群的亲缘关系较远。其中A-2和D-3之间的遗传相似系数最小为0.355,而D-2和D-3之间的遗传相似系数最大为0.575。当遗传相似系数在0.48时,取自四个海拔高度的11个样品可分为3组,Ⅰ组包括A-1,B-2,Ⅱ组包括B-1、C-3、D-2、D-3、B-3、C-1、C-2、D-1,Ⅲ组包括A-2。根据UPGAM聚类图可以看出:西藏野生韭种已经发生了较为强烈的遗传变异。
【Abstract】 The wild Allium hookeri Thwaites from the Tibet is widely distributed in Tibet, which has a special smell and is widely loved by the Tibetan people. However, genetic backgrounds are rarely studied. In this study, 10 pairs of primers of SRAP analysis were used to amplify 11 Tibetan wild Allium hookeri Thwaites.Accordingly, 835 bands were amplified from 10 pairs of primers, of which 826 were polymorphic bands, with a polymorphism proportion of 96.83%. The genetic similarity coefficient between the 11 samples was from 0.355 to 0.575, and the average similarity coefficient was 0.463, indicating that the four populations collected at different altitudes were far related. The genetic similarity coefficient between A-2 and D-3 is 0.355, while the genetic similarity coefficient between D-2 and D-3 is 0.575.When the genetic similarity coefficient of 0.48, from four of the elevation of 11 samples can be divided into three groups, group Ⅰincludes A-1 and B-2, groupⅡ including B-1, C-3, D-2, D-3,B-3, C-1, C-2 andD-1, group Ⅲ includes A-2. According to the UPGAM clustering map, the wild Allium hookeri Thwaites from the Tibet has undergone relatively strong genetic variation.
