3种观赏贝母组培快繁技术

【摘要】 [目的]对3种贝母进行组培快繁。[方法]选择冠花贝母‘鲁布拉’无病无机械损伤中层鳞片的中下部,紫花贝母和弯尖贝母小鳞茎球作为外植体,外植体消毒后进行初代、增殖、生根培养。[结果]75%酒精30 s+10%次氯酸25 min对3种观赏贝母的消毒效果显著;在同样的培养条件下,弯尖贝母、紫花贝母分化率和出苗率高,能够诱导出大量不定芽,增殖系数达4.50和3.98,且出芽整齐,生长良好;冠花贝母多从鳞茎直接诱导产生不定芽,增殖率不高,且生长过程中容易出现内生菌和玻璃化现象。[结论]弯尖贝母、紫花贝母分化率和出苗率高,适合用组织培养技术进行快繁。

【Abstract】 [Objection]The tissue culture rapid propagation technology was used to propagate three kinds of Ornamental Fritillaries. [Method] The explant for tissue culture in the middle scale of F. imperialis‘Rubra Maxima’without disease and mechanical damage,bulb of F. uva-vulpis and F. acmopetala were selected for generation,proliferation,rooting culture after disinfection. [Result] The results showed that the most significant disinfection was 75%alcohol 30 s + 10%sodium hypochlorite 25 min of three kinds of Ornamental Fritillaries. The rate of differentiation and germination was high of F. uva-vulpis and F.acmopetala under the same condition,thus could induce a large number of adventitious bud. The proliferation coefficient was 4.50 and 3.98,and the buds had almost the same length and growth well. The adventitious bud of F. imperialis was induced from bulb,proliferation rate was not high,and prone to endogenous bacteria in the process of growth and vitrification phenomenon. [Conclusion] The rate of differentiation and germination was high of F. uva-vulpis and F. acmopetala,which were suitable for rapid propagation by tissue culture technology.
