粉防己碱对甲状腺癌TP-1细胞的促凋亡作用及其机制
Effect of tetrandrine on apoptosis of thyroid carcinoma TP-1 cells and its mechanism
【摘要】 目的研究粉防己碱对甲状腺癌TP-1细胞增殖、凋亡的影响及其相关机制。方法 TP-1细胞分为4组:对照组和低、中、高剂量实验组。低、中、高剂量实验组分别予以0. 5,10,20μg·m L-1粉防己碱,对照组予以等量0. 9%氯化钠。用Annexin V-FITC/PI双染流式细胞术检测各组TP-1细胞凋亡情况,蛋白免疫印迹法检测细胞YAP、半胱氨酸天冬氨酸蛋白酶-3(Caspase-3)、Survivin、基质金属蛋白酶(MMP)蛋白表达。结果给药48 h后,对照组和低、中、高剂量实验组TP-1细胞细胞凋亡率分别为(9. 02±1. 33)%,(13. 69±1. 58)%,(22. 16±1. 47)%,(36. 85±2. 05)%; YAP蛋白相对表达量分别为1. 56±0. 23,0. 94±0. 11,0. 62±0. 10,0. 53±0. 13; Caspase-3蛋白相对表达量分别为0. 35±0. 15,0. 65±0. 11,0. 85±0. 10,0. 98±0. 06; Survivin蛋白相对表达量分别1. 32±0. 25,0. 89±0. 16,0. 52±0. 07,0. 33±0. 10; MMP蛋白相对表达量分别为0. 98±0. 06,0. 81±0. 03,0. 45±0. 12,0. 23±0. 06;差异均有统计学意义(均P <0. 05)。结论粉防己碱可诱导甲状腺癌TP-1细胞凋亡,其作用机制可能与调控YAP、Caspase-3、Survivin、MMP等凋亡相关蛋白表达相关。
【Abstract】 Objective To investigate the effect of tetrandrine on apoptosis of thyroid carcinoma TP-1 cells and its underlying mechanism.Methods TP-1 cells were divided into 4 groups: control group and experimental-L/-M/-H groups. The experimental-L/-M/-H groups were treated with 0. 5,10,20 μg·m L-1 tetrandrine,while the control group was treated with 0. 9% NaCl. The cell apoptosis rates were detected by Annexin V-FITC/PI double staining flow cytometry,and the expression of YAP,cysteine aspartase 3( Caspase-3),Survivin,matrix metalloproteinase( MMP) protein were detected by Western blot.Results After 48 hours of treatment,there were significant differences between the control group and the the experimental-L/-M/-H group in the apoptosis rates [( 9. 02 ± 1. 33) % vs( 13. 69 ± 1. 58) %,( 22. 16 ± 1. 47) %,( 36. 85 ± 2. 05) % ],the relative expression of YAP protein( 1. 56 ± 0. 23 vs 0. 94 ± 0. 11, 0. 62 ± 0. 10,0. 53 ± 0. 13), the relative expression of Caspase-3 protein( 0. 35 ± 0. 15 vs 0. 65 ± 0. 11,0. 85 ± 0. 10,0. 98 ± 0. 06),the relative expression of Survivin protein( 1. 32 ± 0. 25 vs 0. 89 ± 0. 16,0. 52 ± 0. 07,0. 33 ± 0. 10),the relative expression of MMP protein( 0. 98 ± 0. 06 vs 0. 81 ± 0. 03,0. 45 ± 0. 12,0. 23 ± 0. 06)( P < 0. 05). Conclusion Tetrandrine can induce apoptosis of thyroid carcinoma TP-1 cells by regulating the expression of YAP,Caspase-3,Survivin and MMP.
