【摘要】 目的研究美洲大蠊提取物（PAE）对转化生长因子-β₁（TGF-β₁）体外诱导人胚肺成纤维细胞（HELF）向肌成纤维细胞转化过程中的影响。方法将HELF细胞分为5组:正常组、模型组和低、中、高3个浓度实验组。除正常组外,细胞用TGF-β₁诱导HELF细胞建立体外肺纤维化模型。实验组给予PAE70,80,90μg·m L^-1,模型组和正常组给予含1%胎牛血清（FBS）的培养基。作用24,48,72 h,用比色法检测Ⅰ型胶原蛋白（ColⅠ）和羟脯氨酸（HYP）的水平;以逆转录-聚合酶链反应法检测ColⅠmRNA和结缔组织生长因子（CTGF）mRNA的表达水平。结果 48 h,正常组、模型组和中、高2个浓度实验组的ColⅠ的水平（OD值）分别为0.14±0.00,0.15±0.00,0.14±0.01和0.13±0.00;上述这4组的HYP的含量分别为（2.36±0.14）,（2.81±0.16）,（1.94±0.09）和（1.65±0.22）μg·m L^-1;上述这4组的COLⅠmRNA相对表达量分别为2.46±0.06,2.20±0.03,2.60±0.09和2.86±0.13;上述这4组的CTGF mRNA相对表达量分别为2.59±0.03,2.25±0.09,2.81±0.08和3.10±0.03。上述指标:模型组与正常组比较,差异均有统计学意义（均P<0.01）;中、高2个浓度实验组与模型组比,差异均有统计学意义（均P<0.01）。结论美洲大蠊提取物对TGF-β₁诱导HELF细胞增殖以及向肌成纤维细胞转化过程中具有一定抑制作用。更多还原

【Abstract】 Objective To research the effects of Periplaneta Americana extract（PAE）on the transformation from human embryonic lung fibroblasts（HELF）to myofibroblasts induced by transforming growth factor-β₁（TGF-β₁）in vitro.Methods HELF cells were divided into5 groups:normal group,model group and experimental-L,experimental-M,experimental-H groups.The cells in the normal group did not receive any treatment.Except for the normal group,cells were induced by TGF-β₁to establish the in vitro pulmonary fibrosis model.Experimental groups were treated with 70,80,90μg·m L^-1solution of PAE,model group and normal group were given culture medium containing 1%fetal bovine serum（FBS）.At 24,48 and 72 h,the contents of collagen typeⅠ（ColⅠ）and hydroxyproline（HYP）were detected by colorimetric method.The expression levels of ColⅠmRNA and connective tissue growth factor（CTGF）mRNA were detected by reverse transcriptionpolymerase chain reaction.Results At 48 h,the relative expression levels of ColⅠin normal group,model group,experimental-M group and experimental-H group were 0.14±0.00,0.15±0.00,0.14±0.01,and 0.13±0.00;the HYP contents in the above 4 groups were（2.36±0.14）,（2.81±0.16）,（1.94±0.09）,and（1.65±0.22）μg·m L^-1;the relative expression levels of COLⅠmRNA in the above 4 groups were 2.46±0.06,2.20±0.03,2.60±0.09,and 2.86±0.13;the relative expression levels of CTGF mRNA in the above 4 groups were 2.59±0.03,2.25±0.09,2.81±0.08,and 3.10±0.03.Comparing between model group and normal group,the differences of the factors were significant（all P<0.01）;comparing between two concentration experimental groups and model group,the differences of the factors were significant（all P<0.01）.Conclusion PAE has a certain inhibitory effect on HELFcells proliferation and transformation to myofibroblasts induced by TGF-β₁.更多还原
