山姜素对角叉菜胶诱导的小鼠急性炎症的抗炎作用及机制研究

【摘要】 目的探讨山姜素对角叉菜胶致小鼠急性炎症的抗炎作用及机制。方法按照体重将昆明小鼠随机分为5组:正常组、模型组和低、中、高3个剂量实验组,每组10只。正常组和模型组灌胃0.9%Na Cl,3个剂量实验组分别灌胃山姜素10,20,40 mg·kg-1,每天1次,连续给药7 d。末次给药1 h后,用角叉菜胶复制足肿胀小鼠模型。以酶联免疫吸附法检测肿胀足组织中髓过氧化物酶(MPO)活性、肿瘤坏死因子-α(TNF-α)和白细胞介素-1β(IL-1β)水平,以免疫印迹法检测肿胀足组织中过氧化物酶增殖物活化受体γ(PPARγ)的表达和核因子-кB P65(NF-κB p65)磷酸化水平。结果正常组、模型组和低、中、高3个剂量实验组的MPO分别为(1.15±0.05),(1.90±0.35),(1.71±0.26),(1.54±0.28)和(1.22±0.31)U·g-1;上述这5组的TNF-α分别为(176.3±43.6),(324.9±45.8),(299.3±27.7),(273.8±27.8)和(256.3±26.1)pg·g-1;上述这5组的IL-1β分别为(299.3±10.7),(472.1±39.8),(414.5±26.5),(387.6±10.3)和(346.8±28.0)pg·g-1;上述这5组的PPARγ蛋白表达量(灰度值)分别为0.93±0.05,0.48±0.05,0.58±0.05,0.67±0.06和0.84±0.06;上述这5组的NF-κB p65磷酸化水平(灰度值)分别为0.22±0.04,1.08±0.09,0.83±0.07,0.67±0.05,0.39±0.03。上述指标:模型组与正常组相比,差异均有统计学意义(均P<0.01);3个剂量实验组与模型组相比,差异均有统计学意义(P<0.05,P<0.01)。结论山姜素对角叉菜胶诱导的急性炎症有抗炎作用,其机制可能与PPARγ/NF-κB信号通路有关。

【Abstract】 Objective To explore the anti-inflammatory effects and mechanism of alpinetin on acute inflammation caused by carrageenan in mice.Methods Kunming mice were randomly divided into five groups:normal group(0.9%Na Cl),model group(0.9%Na Cl)and low,middle,high dose experimental groups(alpinetin:10,20,40 mg·kg-1),each group had ten mice,once per day for seven days.One hour after the last administration,paw edema model was established using carrageenan.The levels of the myeloperoxidase(MPO)activity,tumour necrosis factor-α(TNF-α)and interleukin-1β(IL-1β)in paw edema tissue were detected by enzyme-linked immune sorbent assay.The expression of peroxisome proliferator-activated receptorγ(PPARγ)and nuclear factor kappa-B p65(NF-κB p65)in inflammatory paw tissue wereassayed by Western blotting.Results The MPO activities in normal group,model group and low,middle,high dose experimental groups were(1.15±0.05),(1.90±0.35),(1.71±0.26),(1.54±0.28),and(1.22±0.31)U·g-1,respectively;TNF-αlevels in the above 5 groups were(176.3±43.6),(324.9±45.8),(299.3±27.7),(273.8±27.8),and(256.3±26.1)pg·g-1,respectively;IL-1βlevels in the above 5 groups were(299.3±10.7),(472.1±39.8),(414.5±26.5),(387.6±10.3),and(346.8±28.0)pg·g-1,respectively;the expression(grey value)of PPARγin the above 5 groups were 0.93±0.05,0.48±0.05,0.58±0.05,0.67±0.06,and 0.84±0.06,respectively;phosphorylation level(grey value)of NF-κB p65 in the above 5 groups were0.22±0.04,1.08±0.09,0.83±0.07,0.67±0.05,and 0.39±0.03.Comparing between model group and normal group,the differences of the factors were significant(all P<0.01);comparing between the three dose of experimental groups and model group,the differences of the factors were significant(P<0.05,P<0.01).Conclusion Alpinetin displayed significant anti-inflammatory effects on acute inflammation induced by carrageenan,which might be related to the regulation of PPARγ/NF-κB signaling pathway.
