谷胱甘肽对顺铂致雄性小鼠生殖毒性的保护作用
Protective effects of glutathione on reproductive toxicity of cisplatin in mice
【摘要】 目的探讨谷胱甘肽(GSH)对大剂量顺铂致雄性小鼠生殖毒性的保护作用。方法用随机数表法将雄性小鼠随机分为4组:空白组、正常组,模型组和实验组,每组6只。正常组和实验组分别腹腔注射GSH 1200 mg·kg-1,空白组注射等剂量0. 9%Na Cl;连续注射3 d;第3天注射GSH后0. 5 h,模型组和实验组分别一次性腹腔注射顺铂20 mg·kg-1。处死前4 h腹腔注射秋水仙素,给药3 d后处死小鼠。检测小鼠体重、小鼠精子畸形率、精原细胞染色体畸变率和睾丸脏器指数。结果空白组、正常组、模型组和实验组的精子畸形率分别是(6. 54±1. 38)%,(6. 02±1. 04)%,(16. 22±0. 59)%和(12. 18±1. 64)%;上述这4组的精原细胞染色体畸形率分别是(5. 93±1. 39)%,(6. 28±1. 22)%,(14. 93±3. 22)%和(9. 88±1. 62)%;模型组与空白组相比、或者实验组与模型组相比,上述指标的差异均有统计学意义(P <0. 05,P <0. 01)。上述这4组的睾丸系数分别是(0. 31±0. 04)%,(0. 56±0. 06)%,(0. 41±0. 13)%,和(0. 49±0. 09)%;模型组与空白组相比差异有统计学意义(P <0. 05),但是实验组与模型组相比,差异无统计学意义(P> 0. 05)。结论 GSH对顺铂所致雄性小鼠的生殖毒性具有保护作用。
【Abstract】 Objective To investigate the protection of glutathione( GSH) on the male mice which are caused by high dose cisplatin.Methods Male mice were randomly divided into 4 groups: blank group,normal group,model group and experimental group,6 mice in each group. The normal group and the experimental group were intraperitoneally injected with GSH 1200 mg·kg-1 everyday for 3 days,respectively;while the blank group was injected with equal dose of 0. 9% Na Cl. Then the model group and the experimental group were intraperitoneally injected with cisplatin 20 mg·kg-1 respectively. Colchicine was intraperitoneally injected 4 h before the execution. After 3 days administration,the mice were sacrificed,and the body weight,sperm abnormality rate,chromosome aberration rate, and testicular coefficients were measured.Results Sperm abnormality rates in blank group,normal group,model group and experimental group were( 6. 54 ± 1. 38) %,( 6. 02 ± 1. 04) %,( 16. 22 ± 0. 59) %,( 12. 18 ± 1. 64) %,respectively; chromosome aberration rate of spermatogonial cells in the above 4 groups were( 5. 93 ±1. 39) %,( 6. 28 ±1. 22) %,( 14. 93 ±3. 22) %,( 9. 88 ±1. 62) %,respectively; comparison between model group and blank group,the difference of the factors was significantly( all P < 0. 05); comparison between experimental group and model group,the difference of the factors was significantly( all P < 0. 05). The testicular coefficients in the above 4 groups were( 0. 31 ± 0. 04) %,( 0. 56 ± 0. 06) %,( 0. 41 ± 0. 13) % and( 0. 49 ± 0. 09) %,respectively. Comparison between model group and blank group,the difference was significantly( P < 0. 05); comparison between experimental group and model group,the difference was not significantly( P > 0. 05). Conclusion GSH has protective effects on the reproductive toxicity induced by cisplatin in male mice.
