法舒地尔对急性有机磷中毒大鼠的心与肾功能的保护作用

【摘要】 目的研究法舒地尔对急性有机磷中毒大鼠的心、肾功能的保护作用。方法按随机数表法将大鼠分为4组:正常组、模型组和低、高2个剂量实验组,每组18只。在呼吸机支持下,大鼠用有机磷腹腔注射建立急性有机磷中毒大鼠模型。麻醉前,低、高2个剂量实验组腹腔注射盐酸法舒地尔注射液1,10mg·kg-1;模型组和正常组分别腹腔注射0. 9%Na Cl溶液1 m L·kg-1。造模6h后,用酶联免疫吸附法测定血清心肌肌钙蛋白T(c Tn T)、脑钠肽前体(NTPro BNP)水平,用酶法测定血清尿素氮(BUN)、肌酸酐(SCr)水平。结果处置后,正常组、模型组和低、高2个剂量实验组的c Tn T分别为(0. 01±0. 00),(0. 14±0. 02),(0. 09±0. 01)和(0. 03±0. 01) ng·m L-1;这4组的NT-Pro BNP分别为(52. 38±7. 04),(121. 93±15. 37),(103. 24±13. 29),和(61. 97±8. 12)pg·m L-1;这4组的SCr分别为(22. 54±3. 06),(119. 46±15. 47),(87. 68±12. 02)和(51. 67±6. 43) U·L-1;这4组的BUN分别为(5. 82±0. 81),(14. 29±2. 04),(10. 78±1. 25)和(7. 66±1. 07) U·L-1,模型组与正常组比较或者低、高2个剂量实验组与模型组比较,差异均有统计学意义(均P <0. 05)。结论法舒地尔能剂量依赖性的保护急性有机磷中毒大鼠的心功能及肾功能,其作用可能与下调Bax/Bcl-2比值、抑制心肌组织及肾组织的细胞凋亡有关。

【Abstract】 Objective To analyze the protective effects of fasudil on heart and kidney function in rats models with acute organophosphorus pesticide poisoning. Methods According to the random number table,the rats were divided into 4 groups: normal group,model group,experimental-L group and experimental-H group,each with 18 rats. Under the support of the ventilator,the acute organophosphorus pesticide poisoning rats models were established by intraperitoneal injection of organic phosphorus. Before anaesthesia,experimental-L group and experimental-H group were given fasudil hydrochloride injection 1,10 mg·kg-1 respectively,intraperitoneal injection; the model group and normol group were given 0. 9% NaCl solution 1 m L·kg-1,intraperitoneal injection.After 6 h of establishing the model,the levels of serum cardiac troponin T( cTnT),brain natriuretic peptide precursor( NT-ProBNP) were determined by enzyme-linked immunosorbent assay. The urea nitrogen( BUN),creatinine( SCr) were determined by emzyme method. Results After the disposal,the cTnT in normol group,model group, experimental-L group and experimental-H group were respectively( 0. 01 ± 0. 00),( 0. 14 ± 0. 02),( 0. 09 ± 0. 01) and( 0. 03 ± 0. 01) ng·m L-1,NT-ProBNP in the 4 groups were respectively( 52. 38 ± 7. 04),( 121. 93 ± 15. 37),( 103. 24 ± 13. 29) and( 61. 97 ± 8. 12) pg·m L-1; SCr in the 4 groups were respectively( 22. 54 ± 3. 06),( 119. 46 ± 15. 47),( 87. 68 ± 12. 02) and( 51. 67 ± 6. 43) U·L-1; BUN in the 4 groups were respectively( 5. 82 ± 0. 81),( 14. 29 ± 2. 04),( 10. 78 ± 1. 25) and( 7. 66 ± 1. 07) U·L-1. Comparison between model group with normol group or experimental-L group and experimental-H group with model group,the differences of the factors were significant( all P < 0. 05). Conclusion Fasudil can protect heart function and kidney function in the rat with acute organophosphorus poisoning in a dose dependent manner,the effect may be related to the down-regulation of Bax/Bcl-2 ratio,and inhibition of apoptosis in myocardial tissue and renal tissue.
